HMGB1对丙酸睾酮诱导去势大鼠前列腺增生的影响及其初步机制探讨

【摘要】:目的:1.建立大鼠前列腺增生动物模型,筛选丙酸睾酮诱导去势大鼠前列腺增生模型的造模最佳时间点。2.探讨去势大鼠前列腺增生模型注射重组鼠源性HMGB1蛋白对大鼠前列腺的影响及其可能机制。方法:1.48只成年雄性SD大鼠随机分成为6组,每组8只:对照组、去势对照组、7d组(皮下注射丙酸睾酮7天后取材)、14d组、21d组、28d组;测量大鼠体重、前列腺体积、湿重及前列腺指数;采用HE染色法观察前列腺组织病理学改变;免疫组织化学及Western blot法检测大鼠前列腺组织中HMGB1、RAGE、TLR4的表达变化。2.32只成年雄性SD大鼠随机分成为4组,每组8只:21d模型组、低剂量组(去势后往大鼠前列腺组织注射100mg重组鼠源性HMGB1蛋白,同时皮下注射丙酸睾酮21天后取材)、中剂量组(200mg)、高剂量组(400mg);测量大鼠体重、前列腺体积、湿重及前列腺指数;采用HE染色法观察前列腺组织病理学改变;免疫组织化学及Western blot法检测大鼠前列腺组织中HMGB1、RAGE、TLR4的表达变化。结果:1.大鼠去势后前列腺组织萎缩,与对照组及7d组比较,14d组、21d及28d组大鼠前列腺体积、前列腺湿重、前列腺指数均明显增加(P0.05)。且21d组与28d组前列腺体积较14d组明显增大(P0.05),但21d组与28d间各数值无统计学差异。14d组大鼠前列腺上皮细胞和间质组织出现增生改变,21d和28d组大鼠前列腺上皮细胞和间质组织明显增生,且存在炎性细胞浸润。与对照组比较,21d组大鼠前列腺组织HMGB1、RAGE、TLR4的表达升高(P0.05);与对照组比较,28d组大鼠前列腺组织HMGB1、TLR4的表达升高(P0.05)。2.与21d组比较,HMGB1中、高剂量组指数明显增加(P0.05),其余组之间无统计学差异;与21d组比较,HMGB1大剂量组前列腺组织中HMGB1、RAGE、TLR4的表达明显增加(P0.05),其余组之间无统计学差异。结论:1.采用去势后皮下注射丙酸睾酮的方法,21天就可成功构建大鼠前列腺增生模型,且在此过程中伴随HMGB1、RAGE、TLR4的表达升高。2.采用去势大鼠前列腺增生模型中注射中、高剂量重组鼠源性HMGB1蛋白的方法,可使大鼠前列腺指数明显增加,且在此过程中伴随HMGB1、RAGE、TLR4的表达升高。
[Abstract]:Purpose 1. To establish an animal model of prostatic hyperplasia in rats, the optimal time point of prostatic hyperplasia model induced by testosterone propionate in ovariectomized rats was selected. To investigate the effect of recombinant murine HMGB1 protein injection on rat prostate and its possible mechanism in ovariectomized rats with benign prostatic hyperplasia (BPH). Methods one hundred and forty-eight adult male Sprague-Dawley rats were randomly divided into 6 groups: control group (n = 8) and castrated control group (n = 7). Wet weight and prostate index; HE staining method were used to observe the pathological changes of prostate tissue; immunohistochemistry and Western blot method were used to detect the expression of HMGB1 and RAGEGE-TLR4. 2.32 adult male Sprague-Dawley rats were randomly divided into 4 groups. Eight rats in each group were divided into two groups: low dose group (100mg recombinant rat HMGB1 protein was injected into prostate tissue after castration, testosterone propionate was injected subcutaneously for 21 days), middle dose group (200 mg / g), high dose group (400 mg / g), body weight and volume of prostate were measured. The wet weight and prostate index, the pathological changes of prostate tissue were observed by HE staining, and the expression of HMGB1 and RAGEGE-TLR4 in rat prostate tissue was detected by immunohistochemistry and Western blot. The result is 1: 1. Compared with control group and 7 day group, the prostate volume, wet weight and prostatic index of rats in 14 d group and 28 d group were significantly increased compared with control group and 7 d group. The prostate volume of 21 d group and 28 d group was significantly larger than that of 14 d group, but there was no significant difference between 21 d group and 28 d group. There was no significant difference between 21 d group and 28 d group. The interstitial tissue was obviously proliferated. And inflammatory cell infiltration. Compared with the control group, the expression of HMGB1-RAGEGE-TLR4 in the prostate tissue of rats in 21 d group was higher than that in the control group, and the expression of HMGB1 + TLR4 in the prostate tissue of the control group was significantly higher than that in the control group. Compared with the 21D group, the index of HMGB1 increased significantly in high dose group, but there was no statistical difference between the other groups, and the expression of HMGB1 RAGEGE-TLR4 in prostate tissue of high-dose HMGB1 group was significantly higher than that of 21D group, but there was no statistical difference between the other groups. Conclusion 1. The rat model of prostatic hyperplasia was successfully constructed by subcutaneous injection of testosterone propionate 21 days after ovariectomy, and the expression of TLR4 was increased with the increase of HMGB1, RAGEG, TLR4. The high dose recombinant HMGB1 protein in ovariectomized rat model of prostatic hyperplasia could significantly increase the prostatic index, and the expression of HMGB1 / RAGEGE-TLR4 was increased in the process.
