E2F3在肾透明细胞癌中的表达和意义

【摘要】:目的:探讨细胞周期转录因子E2F3在肾透明细胞癌以及对应瘤旁组织中的表达情况,分析E2F3在肾透明细胞癌的发生和恶性进展中所起的作用。 方法:通过实时定量PCR和免疫组化方法检测E2F3在肾透明细胞癌及对应瘤旁组织中mRNA和蛋白的相对表达情况,并分析E2F3的mRNA水平和蛋白水平表达与临床病理资料的相关性。 结果:和对应瘤旁组织相比,E2F3的mRNA水平和蛋白水平在肾透明细胞癌组织中显著下调(mRNA水平0.051±0.073VS1.841±1.551,,P<0.001;蛋白水平0.434±0.109VS0.779±0.103,P<0.001),在TNM分期和临床分期组间有统计学差异(mRNA水平0.018±0.009VS0.184±0.157,P<0.05;蛋白水平0.407±0.115VS0.494±0.063,P<0.05),E2F3的mRNA水平以及蛋白水平在不同年龄、性别组间无统计学差异(P>0.05)。 结论:E2F3表达下调可能在肾透明细胞癌的肿瘤形成中发挥作用,且E2F3表达降低可能促进肾透明细胞癌的恶性进展。
[Abstract]:Aim: to investigate the expression of cell cycle transcription factor E2F3 in renal clear cell carcinoma and its adjacent tissues, and to analyze the role of E2F3 in the development and malignant progression of renal clear cell carcinoma. Methods: the relative expression of E2F3 mRNA and protein in renal clear cell carcinoma and its adjacent tissues were detected by real-time quantitative PCR and immunohistochemistry, and the correlation between the expression of E2F3 mRNA and protein and clinicopathological data was analyzed. Results: the mRNA and protein levels of E2F3 were significantly decreased in renal clear cell carcinoma (0.051 卤0.073 vs 1.841 卤1.551, P < 0.001; protein level 0.434 卤0.109 vs 0.779 卤0.103, P < 0.001), and there was a significant difference between TNM staging and clinical staging (0.018 卤0.009 vs 0.184 卤0.157, P < 0.05). The protein level of E2F3 was 0.407 卤0.115 vs 0.494 卤0.063 (P < 0. 05). There was no significant difference in the level of E2F3 mRNA and protein between different age and sex groups (P > 0. 05). Conclusion the down-regulation of the expression of E2F3 may play a role in tumor formation of renal clear cell carcinoma, and the decreased expression of E2F3 may promote the malignant progression of renal clear cell carcinoma.
