【摘要】:目的制备一种用于对人前列腺癌PC3细胞进行体外检测的粒细胞靶向介导的磁性-荧光纳米探针。方法制备以Fe_3O_4为核、表面包被SiO_2和异硫氰酸罗丹明的具有磁性且携带红色荧光的纳米材料,将其与正常人外周血粒细胞按不同比例混合,分别孵育不同时间以检测纳米材料对粒细胞的毒性。选择最佳比例将纳米材料和粒细胞体外结合,得到粒细胞靶向介导的磁性-荧光纳米探针;将PC3细胞与正常人全血细胞分别以不同比例混合,加入上述探针,于荧光显微镜下观察探针的靶向情况。结果在实验所用的浓度和作用时间范围内,制备的纳米探针对粒细胞存活率无明显影响。镜下可见纳米探针在PC3细胞周围富集,形成"花瓣样"结构,而全血细胞周围无探针富集。结论本研究制备的磁性-荧光纳米探针不会对粒细胞本身产生明显毒性,利用粒细胞对肿瘤细胞的生物靶向作用可使纳米探针有效地靶向检测肿瘤细胞。
[Abstract]:Objective to prepare a magnetic fluorescent nano probe mediated by granulocyte targeting for human prostate cancer PC3 cells in vitro, and to prepare nano materials with magnetic and red fluorescence with SiO_2 and isothiocyanate Luo Danming as the core of Fe_3O_4 and mixed with normal human peripheral blood granulocytes in a different proportion. In order to detect the toxicity of nanomaterials to granulocytes at different time, the optimum proportion was chosen to combine the nanomaterial and granulocyte in vitro, and the magnetic fluorescent nano probe mediated by granulocyte targeting was obtained, and the PC3 cells were mixed with the normal human whole blood cells in different proportions, and the probe was observed under the fluorescence microscope. The results showed that there was no obvious effect on the survival rate of granulocytes in the concentration and time range used in the experiment. The nano probe was enriched around PC3 cells and formed a petal like structure under the microscope. There was no probe enrichment around the whole blood cells. It has obvious toxicity to granulocytes. The biological targeting effect of granulocytes on tumor cells can make nanomaterials effectively detect tumor cells.
