【摘要】:目的 Tamm-Horsfall蛋白(THP)在肾结石形成过程中可能起一定作用。文中旨在通过建立钙化性纳米微粒(CNPs)大鼠肾结石模型,测定大鼠尿液中THP的变化、肾组织病理改变及钙盐晶体形成情况,对THP在肾结石形成过程中的作用机制进行初步研究。方法取泌尿系患者的结石标本,建立CNPs致大鼠肾结石模型,制成CNPs混悬液。将48只SD大鼠随机分为实验组(注射CNPs混悬液)和对照组(注射等渗盐水),于注射后3 h、6 h、12 h、24 h、1 w、2 w、4 w和8 w收集大鼠尿液,ELISA检测尿液中THP的含量,取肾组织行HE染色观察细胞的病理改变和对钙盐晶体形成情况进行评分。结果实验组24h、1w、2w、4w、8w THP含量[(166.03±3.02)、(173.50±1.78)、(174.55±2.05)、(176.54±2.45)、(177.11±1.76)pg/mL]较3 h[(165.89±2.23)pg/mL]明显增加(P0.05);亦较对照组24 h、1 w、2 w、4 w、8 w[(157.65±2.22)、(156.54±1.43)、(159.45±3.21)、(158.63±2.98)、(157.33±2.05)pg/mL]明显升高(P0.05)。与实验组6 h钙盐晶体评分(1.0分)比较,3、6、12、24 h的钙盐晶体评分(均为2.0分)增加,2w后钙盐晶体评分增加显著(3.0分),8w评分最高(6.7分),差异有统计学意义(P0.05)。钙盐晶体的评分与THP含量呈正相关(r=0.843,P0.05)。结论尿液中的THP可以促进钙盐晶体的聚集成核,有利于肾结石的形成。
[Abstract]:Objective Tamm-Horsfall protein (THP) may play a role in the formation of renal calculi. The aim of this study was to establish a rat model of calcified nanoparticles (CNPs), to determine the changes of THP in urine, the pathological changes of renal tissue and the formation of calcium crystals, and to study the mechanism of THP in the formation of renal calculi. Methods Nephrolithiasis model was established in rats with CNPs and the suspension of CNPs was made. Forty-eight Sprague-Dawley rats were randomly divided into two groups: experimental group (injected with CNPs suspension) and control group (injected with isoosmotic saline). THP levels in urine of rats were collected at 3 h, 6 h, 12 h, 24 h, 1 week, 2 weeks, 4 weeks and 8 weeks after injection.
