hAMSCs移植对鱼胆中毒大鼠肾组织中CD80分子表达的影响
[Abstract]:Aim: to study the changes of CD80 expression in renal tissue of rats with acute tubular necrosis (acute tubular necrosis) treated with human amniotic mesenchymal stem cells (human amniotic mesenchymal stem cells) and to explore the mechanism of transplantation of human amniotic mesenchymal stem cells (hAMSCs) in the treatment of fish bile duct poisoning rats. Methods: the rat model of ATN poisoning was prepared by fish gall instillation in the early stage of the study group. The rats were treated by tail vein injection of hAMSCs on day 3 and day 7, and the paraffin tissues of renal tissue on day 14 were collected. The rats were divided into control group (normal rats), model group (fish bile poisoning rats) and transplantation group (hAMSCs transplantation treated rats). The integrated optical density (IOD) was measured by collecting pictures, and semi-quantitative analysis was carried out. Results: (1) hAMSCs were transplanted on day 3 and day 7 and day 14, respectively. There was no significant difference in the expression of CD80 between the control group and the transplantation group (P0.05). The expression of CD80 in the model group was higher on the 3rd day than on the 7th day (P0.05), and on the 7th day, there was no significant difference between the control group and the transplantation group (P0.05) on the 3rd day after transplantation (P0.05), and on the 7th day after transplantation, there was no significant difference in the expression of CD80 between the control group and the transplantation group (P0.05). The expression of CD80 in renal tubular epithelial cells in the model group was the lowest (P0.05), but there was no significant difference between the model group and the control group on the 14th day (P0.05). The expression of CD80 on the 3rd day (P0.05) in the transplantation group was the highest (P0.05), and there was no significant difference (P0.05) on the 14th day (P0.05) in the 3rd day and the 14th day in the transplantation group (P0.05). CD80 expression was not found in renal tubular epithelial cells in the medullary region of the control group (P0.05). The expression of CD80 in the medullary area of the model group was higher on the 3rd day than on the 7th day (P0.05), and decreased significantly on the 7th day, but higher than that on the 14th day (P0.05) on the 3rd day after transplantation of hAMSCs. The expression of CD80 in the model group was higher than that in the transplantation group (P0.05), but it was higher in the transplantation group than that in the control group (P0.05). Conclusion the expression of CD80 in glomeruli and renal tubular epithelial cells may be inhibited by the transplantation of 10% hAMSCs in ATN rats, and then the activation of T cells and inflammation can be alleviated.
