CircCD44-360与circCD44-594在膀胱癌中的表达及临床意义

【摘要】:目的:本研究就circCD44-360(hsa_circ_0021727)与circCD44-594(hsa_circ_0021728)在膀胱癌及癌旁组织中的表达情况,及它们对CD44基因表达的影响和相关性分析进行研究,旨在为膀胱癌的诊断和靶向治疗提供新的方向。方法:收集膀胱癌及癌旁组织的临床标本,处理标本后提取膀胱癌及癌旁组织的总RNA,反转录获取目的RNA,进行实时定量PCR,检测circCD44-360与circCD44-594的表达差异,并分析其分别同亲本基因CD44的信使RNA(messenger RNA,mRNA)的关系进而找出其调节机制。结果:1 CircCD44-360与circCD44-594在膀胱癌组织的表达均上调。PCR结果显示,与癌旁组织相比,circCD44-360与circCD44-594在膀胱癌组织中的表达水平显著升高。说明circCD44-360与circCD44-594在膀胱癌组织中的表达水平均显著上调。2 CD44mRNA在膀胱癌组织的表达上调。PCR结果显示,与癌旁组织相比,CD44mRNA在膀胱癌组织中的表达水平显著升高。说明CD44mRNA在膀胱癌组织中的表达水平均显著上调。3 CircCD44-360与circCD44-594均同其亲本基因CD44的表达呈线性正相关关系。利用PCR结果进行统计学分析,circCD44-360与其亲本基因CD44的Pearson's correlation线性相关关系,R=0.5702,P=0.0087,呈正相关。另一个环状circCD44-594与其亲本基因CD44的Pearson's correlation线性相关关系,R=0.4474,P=0.0420,呈正相关。结论:1 CircCD44-360在膀胱癌组织中表达上调。2 CircCD44-594在膀胱癌组织中表达上调。3在膀胱癌中,circCD44-360、circCD44-594和CD44的表达呈正相关关系,它们可能通过调节CD44基因的表达促进膀胱癌的发生和发展。
