转移性肾细胞癌中RSK4、CD44及MMP-9的表达及意义

【摘要】:目的检测核糖体S6蛋白激酶4(ribosomal S6 protein kinase 4,RSK4)、CD44和MMP-9在原发性肾细胞癌(primary renal cell carcinoma,pRCC)及转移性肾细胞癌(metastatic renal cell carcinoma,mRCC)中的表达,探讨三者表达水平及与临床病理特征的相关性。方法采用免疫组化En Vision法检测52例pRCC及48例mRCC中RSK4、MMP-9和CD44蛋白的表达,并分析三者表达的相关性及与临床病理特征的关系。结果 48例mRCC中RSK4、CD44及MMP-9的阳性率分别为75%(36/48)、68.75%(33/48)及91.7%(44/48),而三者在52例pRCC中的阳性率分别为44.2%(23/52)、34.6%(18/52)及69.2%(36/52)。RSK4、CD44及MMP-9在mRCC中的表达高于pRCC(PRSK4=0.002;PMMP-9=0.002;PCD44=0.001);RSK4、CD44及MMP-9表达与mRCC患者年龄、性别和肿瘤Fuhrman分级、转移部位之间未见明确相关性(P0.05)。在mRCC中,三者之间的表达呈正相关(P=0.008);两两相关分析发现RSK4与CD44(P=0.019)、MMP-9与CD44(P=0.05)的表达之间存在正相关,而RSK4与MMP-9(P=1.00)未见明确相关性。结论 RSK4、CD44及MMP-9在mRCC中的表达高于pRCC,提示三者可能介导肾细胞癌(renal cell carcinoma,RCC)的转移,而其具体作用机制尚有待进一步研究。
