Galanin及Galanin2型受体在前列腺组织中的表达及其临床意义
[Abstract]:Objective: to investigate the expression and clinical significance of Galanin and Galanin2 receptor (Ga1R2) in benign and malignant prostate tissues. Methods: 50 cases of benign prostatic hyperplasia (BPH) and 50 cases of (PCa) without metastasis and 30 cases of (BM) with bone metastasis were collected. The expression of Galanin and Ga1R2 in these tissues was observed by immunohistochemical method and its clinical significance was analyzed. We selected human prostatic hyperplasia cell line BPH-I, prostate cancer cell line DU145 as the research object. After two groups of cells were cultured, the total cell protein was extracted, and then the expression of Galanin and Ga1R2 protein in the two groups were detected by Western blot. Results: immunohistochemical results showed that the positive expression of Galanin protein in BPH group (9 cases) was 34 cases in BM group (34 cases) and in BPH group (40 cases), 7 cases (7 cases) in BPH group, 2 cases in BM group. The results showed that the positive rate of Galanin expression in PCa group and BM group was significantly higher than that in BPH group (蠂 2 = 25.50 and 32.75, respectively, P0.01). There was no significant difference between PCa group and BM group (蠂 2 / 2.28 P 0.05). The positive rate of GalR2 expression in BPH group was significantly higher than that in PCa group and BM group (蠂 ~ 2 = 43.72 and 40.43, respectively, P0.01). There was no significant difference between PCa group and BM group (蠂 ~ (2) 1.01). In PCa group and BM group, the expression of Galanin and Ga1R2 protein was negatively correlated (rs=-0.55,P0.01;rs=-0.77,P0.01). Western blotting showed that the expression of Galanin protein in BPH-1 cells decreased significantly, but increased in DU145 cells, on the contrary, the expression of Galanin protein in BPH-I cells increased. The expression in DU145 cells was significantly decreased. The results were consistent with the immunohistochemical results. Conclusion the positive expression rate of Galanin in prostate cancer tissues is high, suggesting that Galanin may be involved in the pathogenesis of prostate cancer, while the protein expression level of GalR2 in prostate cancer tissue is decreased. The expression of GalR2 and Galanin protein was negatively correlated with immunohistochemical results, suggesting that Galanin may play a biological role in prostate cancer by combining with two receptors other than Ga1R2. It is also suggested that the combined detection of Galanin and Ga1R2 protein levels has a good clinical significance in the diagnosis of benign and malignant prostate diseases.
