WEE1在肾透明细胞癌中的表达及意义

【摘要】:目的:研究W E E1基因在肾透明细胞癌(RCCC)中的表达和意义,分析W E E1基因与肾透明细胞癌的临床分期、病理分级及淋巴转移的关系,为肾细胞癌(主要为透明细胞癌)的诊断治疗及预后提供理论指导。 方法:应用免疫组织化学、real time PCR和Western blot法检测32例肾透明细胞癌和25例癌旁正常肾组织中W E E1的表达情况,并将W E E1转录及表达水平与RCCC的生物学效应进行相关分析;采用SPSS14.0软件统计学分析实验数据,设定P0.05为差异具有统计学意义。 结果:32例肾透明细胞癌组织中W E E1的阳性表达为28例(87.5%),阴性表达为4例(12.5%),25例正常肾组织中W E E1阳性表达为2例(8.0%),阴性表达为23例(92%)。两组间比较,差异具有统计学意义(P0.01);肾透明细胞癌组织和正常组织W E E1表达CT值分别为13.7±3.7和4.5±2.0,两组比较有统计学差异(P0.01),肾癌组织的W E E l表达显著大于正常组织中的W E E1表达。Western Blot检测结果显示,肾癌组织和正常组织W E E1表达相对灰度比值分别为5.2±1.5和0.6±0.37,两组比较有统计学差异(P0.01),肾癌组织的W E E1表达显著大于正常组织中的W E E1蛋白表达。W E E1表达水平与临床分期密切相关,Ⅰ、Ⅱ和Ⅲ-Ⅳ期相对灰度比值分别为3.1±0.7,4.7±0.9和6.7±0.9,其Ⅱ和Ⅲ-Ⅳ期肾癌分别是Ⅰ期的1.5倍和2.2倍。(P0.05)。W EE1表达水平与组织分级密切相关,高分化、中分化和低分化肾癌灰度比值分别为3.1±0.5,4.4±0.5和6.4±0.9,中分化、低分化肾癌灰度比值分别是高分化肾癌的1.75倍、1.80倍(P0.05)。W E E1表达水平与淋巴转移也密切相关,淋巴无转移组和转移组W E E1灰度比值分别为4.3±1.1和6.8±0.6,淋巴转移组W E E1灰度比值是无转移组肾癌的1.6倍(P0.05)。W E E l表达和淋巴转移呈正相关,相关系数为0.805(P0.01)。 结论:1、W E E1在肾透明细胞癌转录与表达明显高于对照组,对照组中W E E1低表达。2、W E E1的表达可能与肾透明细胞癌的临床分期、病理分级、淋巴转移相关。当患者处于Ⅲ-Ⅳ期时,WEE1肾细胞癌中的表达量比处于Ⅰ、Ⅱ期患者肾细胞癌中表达高,中、低分化中的表达显著大于高分化,在有淋巴转移的肾透明细胞癌中的表达明显高于无淋巴转移的肾透明细胞癌。3、W E E1有望成为判断肾透明细胞癌转移和预后等生物学行为的重要标记物,将有可能成为肾透明细胞癌治疗的新靶点。
[Abstract]:Objective: to study the expression and significance of WEE1 gene in (RCCC) of renal clear cell carcinoma (RCC), and to analyze the relationship between WE1 gene and clinical stage, pathological grade and lymphatic metastasis of renal clear cell carcinoma (RCC). To provide theoretical guidance for the diagnosis, treatment and prognosis of renal cell carcinoma (mainly clear cell carcinoma). The difference between the two groups was statistically significant (P0.01). The CT expression of WEE1 in renal clear cell carcinoma and normal tissues were 13.7 卤3.7 and 4.5 卤2.0, respectively. There was a significant difference between the two groups (P0.01). The expression of W E E l in renal cell carcinoma was significantly higher than that in normal tissues.
