芒果苷改善高果糖所致自发性高血压大鼠肾间质纤维沉积的可能机制

【摘要】:目的探讨芒果苷(mangiferin,MA)对高果糖引起的自发性高血压大鼠(spontaneously hypertensive rats,SHRs)肾间质纤维沉积的影响及其可能的机制。方法 24只SHRs按随机单位组设计分组法分为对照组、果糖组、果糖+低剂量芒果苷(5 mg/kg)组和果糖+高剂量芒果苷(15 mg/kg)组。实验周期为7周,7周末处死大鼠,取血浆、肾脏组织。检测血浆葡萄糖、甘油三酯(TG)、总胆固醇(TC)、游离脂肪酸(non-esterified fatty acid,NEFA)和肾组织甘油三酯含量,Masson染色后半定量评估肾间质纤维沉积的程度,Real-time PCR检测肾组织单核细胞趋化蛋白-1(monocyte chemoattractant protein-1,MCP-1)、CD68、α-平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)、Ⅳ型胶原蛋白α-Ⅳ链[collagen alpha-4(Ⅳ)chain,col4a4]、转化生长因子-β(transforming growth factor-beta,TGF-β)、纤溶酶原激活物抑制剂-1(plasminogen activator inhibitor-1,PAI-1)、尿激酶纤溶酶原激活物(urokinase plasminogen activator,u PA)mRNA的表达,免疫组化检测肾脏u PA、PAI-1蛋白表达水平。结果与对照组比较,果糖组SHRs肾间质胶原纤维沉积率显著增加(P0.01),而芒果苷(15 mg/kg)明显减轻果糖引起的SHRs肾间质胶原纤维沉积(P0.05)。各组肾脏甘油三酯含量没有变化;肾组织MCP-1、CD68、α-SMA、col4a4、TGF-βmRNA也没有明显变化,但芒果苷(15 mg/kg)明显逆转果糖引起的u PA/PAI-1 mRNA比值的降低(P0.05),同时降低高果糖引起PAI-1蛋白表达水平升高。结论芒果苷能改善高果糖所致SHRs肾脏间质纤维沉积,其机制可能与调控肾组织PAI-1、u PA有关。
