海昆肾喜胶囊和百令胶囊对尿酸性肾病的影响

【摘要】:目的:探讨海昆肾喜胶囊及百令胶囊对高尿酸肾病的防治作用及其可能的作用机制。方法:6周龄SPF级雄性Wistar大鼠93只,普通饲料适应性喂养1周后,随机分为正常组、高尿酸血症组、海昆肾喜干预组、百令胶囊干预组。高尿酸组给予腺嘌呤灌胃50mg/(kg.d),高嘌呤饲料喂养,同时给予氧嗪酸钾乳悬液4ml/(kg. d)分两次皮下注射;干预组大鼠在上述饲养给药的基础上同时给予相应的药物灌胃治疗;正常组大鼠给予普通饲料饲养作为正常对照。在实验的第2、4、6周每组随机选取6只大鼠,内眦静脉取血,检测尿酸(UA)、尿素氮(BUN)、肌酐(CR)及肌酐清除率(Ccr);收集大鼠24h尿液,测定24h尿微量白蛋白定量;同时每组处死2只大鼠,测定左肾质量指数,取肾脏组织行HE, MASSON染色,免疫组化测定白介素1-β(IL-1β)、转化生长因子-β(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)的表达。结果:与正常组比较,高尿酸血症组在第2、4、6周UA、BUN, CR,24小时尿蛋白,左肾质量分数升高,Ccr降低,结果有统计学意义(p0.01)。与高尿酸血症组比较,海昆肾喜干预组在第2,4,6周CR,Ccr,24小时尿蛋白有明显降低(p0.01),第6周BUN,左肾质量分数有明显改善(p0.01);百令胶囊干预组在第2、4、6周Ccr,24小时尿蛋白有明显降低,第2、6周BUN有明显降低,第4、6周CR,左肾质量分数有明显改善(p0.01)。与正常组相比,高尿酸血症组在第2、4、6周HE及MASSON染色提示大鼠肾间质胶质纤维增加,肾间质组织表达TGF-β1,IL-1β,α-SMA明显增加(p0.01)。与高尿酸血症组比较,海昆肾喜干预组在第2、4、6周,肾间质沉淀及纤维化减轻,TGF-β1,IL-1β,α-SMA表达明显减少(p0.01);百令胶囊干预组在第4、6周,肾间质沉淀及纤维化减轻,TGF-β1,IL-1β,α-SMA表达明显减少(p0.01)。 结论:海昆肾喜胶囊及白令胶囊能改善高尿酸肾病大鼠肾脏功能,减少肾间质TGF-β1,IL-1β,α-SMA的表达,减轻肾间质纤维化。
[Abstract]:Objective: to investigate the preventive and therapeutic effects of Haikunshenxi capsule and bailing capsule on hyperuric acid nephropathy and its possible mechanism. Methods: 93 SPF grade male Wistar rats aged 6 weeks were randomly divided into normal group, hyperuricemia group, Hishenkunxi intervention group and bailing capsule intervention group. In hyperuricemia group, 50mg/ (kg.d) was given orally, and hyperpurine was fed on diet and 4ml/ (kg.) was administered with potassium oxazinate milk suspension. D) two times of subcutaneous injection; intervention group rats on the basis of the above feeding and administration of the corresponding drug infusion treatment; normal group rats were fed with normal diet as normal control. Six rats in each group were randomly selected at the 2nd week and 4th week of the experiment. The blood was collected from the inner canthus vein. The uric acid (UA), urea nitrogen (BUN), creatinine (CR) and creatinine clearance rate (Ccr);) were collected from 24 hours urine of the rats. At the same time, two rats were killed in each group. The left renal mass index was measured. The renal tissues were stained with HE, MASSON, and interleukin-1 尾 (IL-1 尾) and transforming growth factor- 尾 (TGF- 尾 1) were detected by immunohistochemistry. Expression of 伪-smooth muscle actin (伪-SMA). Results: compared with the normal group, the urine protein, left renal mass fraction and Ccr were significantly increased in hyperuricemia group at the 6th week of the 2nd week (p0.01). Compared with the hyperuricemia group, the urine protein of CR,Ccr,24 in the intervention group was significantly decreased at the 2nd week (p0.01), and the left renal mass fraction of the BUN, was significantly improved at the 6th week (p0.01). In the intervention group, urinary protein decreased significantly at the 2nd week, the BUN decreased at the 6th week, and the left renal mass fraction of CR, improved significantly at the 4th week (p0.01). Compared with normal group, HE and MASSON staining in hyperuricemia group at the 2nd week and 4th week showed that the expression of TGF- 尾 1 and IL-1 尾 and 伪 -SMA in renal interstitial tissue was significantly increased (p0.01). Compared with the hyperuricemia group, the renal interstitial sedimentation and fibrosis were alleviated and the expression of IL-1 尾, 伪-SMA decreased significantly in the Hishenhei intervention group at the 4th week (p0.01). In the intervention group, the renal interstitial sedimentation and fibrosis were alleviated, and the expression of IL-1 尾, 伪-SMA of TGF- 尾 1 was significantly decreased in the intervention group at the 4th week (p0.01). Conclusion: Haikunshenxi capsule and Bering capsule can improve the renal function, reduce the expression of TGF- 尾 1 and IL-1 尾, 伪-SMA and alleviate renal interstitial fibrosis in rats with hyperuricemia nephropathy.
