半乳糖凝集素-7在人肾移植排斥反应中的初步研究
[Abstract]:Galactose lectin-7 (Galactose lectin,Galectin-7) is a kind of proteins that can selectively recognize the structure of sugar and bind to it noncovalently. It exists in a wide range of organisms from low-level organisms to mammals and regulates the growth of tissues. Intercellular adhesion, apoptosis, tumorigenesis and immune response play an important role in biology. The purpose of this study is to study the role and significance of galectin-7 in immune rejection after human renal transplantation and its relationship with allograft rejection, so as to provide a new way of thinking for the diagnosis and treatment of renal transplantation rejection. From December 2011 to October 2013, 65 patients underwent renal allograft in the 309th Hospital of the Chinese people's Liberation Army (PLA), according to clinical biochemical indicators, The patients were divided into acute rejection group (n = 34) and chronic rejection group (n = 34) by HE and Masson staining (diagnostic criteria of Banff2005 renal transplantation rejection biopsy). There were 34 patients with acute rejection, 16 males and 18 females, with an average age of (43 卤16.3) years old (22 卤57 years old) with a mean age of (43 卤16.3) years. 31 cases of chronic rejection, 17 males and 14 females, aged 27 to 54 years, mean age (41 卤17.8) years. In addition, 30 healthy donors (18 males and 12 females, with an average age of (34 卤13.1) years) were selected as the control group. Immunohistochemical staining and Western Blot method were used to detect the expression and distribution of galectin-7 in renal tissue and the difference of galectin-7 expression in serum of each group. The results showed that the structure of glomeruli and tubules was clear and no obvious abnormality was observed in the normal group. The expression of galectin-7 was found in the lumen of renal tubules and arterial smooth muscle cells, but no expression was found in other parts of the renal tubular lumen and artery smooth muscle cells. In acute rejection group, Mesangial matrix proliferation, renal tubule dermatitis, a large number of lymphocyte infiltration in renal interstitial, no fibrosis in renal interstitial, no atrophy of renal tubule, and the expression of galectin-7 in tubular cavity were observed in most of the cases under light microscope, and there was no fibrosis in renal interstitial tissue and no atrophy of renal tubule in acute rejection group. There was no expression of galectin-7 in some of the lumen. In the chronic rejection group, Mesangial hyperplasia of glomeruli, fibrosis of renal interstitial and matrix, atrophy of renal tubule, no obvious supratubular dermatitis, arterial intimal hyperplasia and a small amount of lymphocyte infiltration were observed in the chronic rejection group. The high expression of galectin-7 in renal tubular epithelial cells was observed in some cases. The gray value of normal control group was (0.32 卤0.15), that of acute rejection group was (0.95 卤0.11), and that of acute rejection group was (0.95 卤0.11). The gray value of chronic rejection group was (1.32 卤0.19). SPSS20.0 software was used to carry out statistical component data, mean 卤standard deviation (), data were processed by one-way ANOVA, and correlation analysis was made among the indexes. The acute rejection group was higher than the normal control group, P 0.05, the difference was statistically significant. The chronic rejection group was higher than the normal control group (P 0.05), and the chronic rejection group was higher than the acute rejection group (P 0.05). In this study, we concluded that galectin-7 may play an important role in renal allograft rejection, and may be a potential target for the treatment of renal allograft rejection.
