辣椒素对膀胱癌细胞侵袭转移的影响

【摘要】:目的探讨辣椒素对膀胱癌细胞系253J与RT112侵袭转移的影响。方法以0、100、200μmol/L辣椒素分别作用于人膀胱癌253J与RT112细胞0、12、24 h,采用MTT法检测253J与RT112细胞活力,划痕实验检测253J与RT112细胞迁移能力,Transwell法检测253J与RT112细胞侵袭能力。Western blotting法检测上皮间充质转化相关蛋白(E-钙黏蛋白,N-钙黏蛋白,波形蛋白)和Nrf2相关蛋白的表达水平。结果与0μmol/L比较,在100、200μmol/L辣椒素作用下,253J与RT112各组细胞活性降低(P0.01),侵袭能力下降(P0.01),迁移能力也下降(253J:P0.01;RT112:P=0.026或P=0.014)。细胞E-钙黏蛋白、C-nrf2表达上升,而波形蛋白与N-nrf2蛋白表达减弱,并呈剂量-时间依赖性。结论辣椒素可抑制人膀胱癌细胞的侵袭与转移,该作用可能与Nrf2的调节有关。
[Abstract]:Objective to investigate the effect of capsaicin on invasion and metastasis of bladder cancer cell line 253J and RT112. Methods Human bladder cancer cell line 253J and RT112 cells were treated with 0100200渭 mol / L capsaicin for 0,12,24 h, respectively. The viability of 253J and RT112 cells was detected by MTT assay, and the migration ability of 253J and RT112 cells was detected by scratch assay. The invasiveness of 253J and RT112 cells was detected by Transwell, and the expression of E-cadherin (N-cadherin, vimentin) and Nrf2-associated protein were detected by Western blotting. Results compared with 0 渭 mol / L capsaicin, the cell activity, invasion ability and migration ability of 253J and RT112 groups were decreased (P0.01), and the migration ability was also decreased (253J / mol / L capsaicin 0.026 or 0.014). The expression of E-cadherin and C-nrf2 increased, but the expression of vimentin and N-nrf2 decreased in a dose-time dependent manner. Conclusion capsaicin can inhibit the invasion and metastasis of human bladder cancer cells, which may be related to the regulation of Nrf2.
