高糖对大鼠膀胱平滑肌细胞中钙库操控的钙离子通道的作用研究

【摘要】:目的:探索高糖对大鼠膀胱平滑肌细胞(bladder smooth muscle cells,BSMCs)中钙库操控的钙离子通道(store-operated calcium channel,SOCC)的影响。通过高糖处理BSMCs,模拟糖尿病膀胱(diabetes cystopathy,DCP)体外环境,探究BSMCs中SOCC的相关变化,为DCP的病因学及其治疗提供新的思路。方法:使用Ⅱ型胶原酶分离S-D大鼠BSMCs,并用α-SMA进行免疫荧光鉴定。分离的原代BSMCs进行培养传代。将传至第3-5代的BSMCs同步化后根据培养基成分及浓度分为:正常葡萄糖浓度组(葡萄糖5.5mmol/L,NG组),甘露醇高渗组(葡萄糖5.5mmol/L+甘露醇19.5mmol/L,OC组),高葡萄糖浓度组(葡萄糖25mmol/L,HG组)三组,于37℃、5%CO2的孵箱中培养2.5天。各组BSMCs负载Fluo-4AM,通过激光共聚焦显微镜观察记录三组BSMCs激活(CPA,10umol/L)和抑制SOCC(SKF-96365,10umol/L)后细胞内荧光强度值,计算其荧光强度值(F1)与背景荧光强度值(F0)的比值(F1/F0)来比较各组间钙离子浓度的变化。通过RT-qPCR法和Western blot法检测三组BSMCs中SOCC相关蛋白STIM1和Orai1的表达情况。结果:经过α-SMA免疫荧光鉴定证实成功分离培养出膀胱平滑肌细胞。HG组加入SOCC激动剂CPA后较NG组、OC组钙离子浓度的显著增加(2.105±0.105,P0.05),加入SOCC抑制剂SKF-96365后较NG组、OC组钙离子浓度的显著降低(1.610±0.109,P0.05),NG组与OC组之间无明显差异(P0.05);HG组BSMCs中STIM1和Orai1的mRNA表达水平以及蛋白表达水平明显高于NG组、OC组(P0.05),NG组与OC组之间无明显差异(P0.05)。结论:经过2.5天高糖作用下能激活大鼠膀胱平滑肌细胞的SOCC,可能参与组成细胞外钙离子内流部分,从而参与相应的病理生理机制。另外,在高糖作用下,BSMCs中SOCC组成分子STIM1和Orai1表达水平发生改变可能为DCP的病理机制提供体外实验依据,从而为DCP病因学研究提供新的思路。但高糖对SOCC的作用机制尚不清楚,仍需后续研究。
