LRPPRC对前列腺癌细胞凋亡的影响

【摘要】:目的:分析富含亮氨酸的三角状五肽重复结构蛋白(LRPPRC)和抗凋亡蛋白Bcl-2在前列腺癌组织的表达及LRPPRC对前列腺癌细胞凋亡的影响。方法:应用免疫组化法检测198例前列腺癌组织中LRPPRC和Bcl-2的表达;分析LRPPRC和Bcl-2表达的相关性;分析LRPPRC和Bcl-2表达与预后的关系;应用si RNA LRPPRC转染前列腺癌细胞下调LRPPRC水平,观察对细胞增殖、凋亡和Bcl-2、Bax、caspase-3蛋白表达的影响。结果:前列腺癌和前列腺增生组织LRPPRC的表达率分别为61.6%(122/198)和18.0%(18/100)(P0.001);Bcl-2的表达率分别为49.5%(98/198)和45.0%(45/100)(P=0.539)。LRPPRC和Bcl-2的表达有显著正相关性(r=0.179,P=0.012)。LRPPRC和Bcl-2同时高表达的患者无疾病进展生存和总生存期均显著降低(P0.001)。下调LRPPRC表达可抑制前列腺癌细胞增殖,促进前列腺癌细胞凋亡,降低Bcl-2蛋白的表达,使caspase-3活化增多,对Bax表达无显著影响。结论:前列腺癌组织LRPPRC表达显著增高;LRPPRC与抑制凋亡有关;LRPPRC在前列腺癌中的作用机制与抗凋亡蛋白Bcl-2有关。
[Abstract]:Aim: to analyze the expression of trigonometric pentapeptide repeat protein (LRPPRC) and anti-apoptosis protein Bcl-2 rich in Leucine in prostate cancer and the effect of LRPPRC on apoptosis of prostate cancer cells. Methods: the expression of LRPPRC and Bcl-2 in 198 cases of prostate cancer was detected by immunohistochemistry, the correlation between the expression of LRPPRC and Bcl-2 was analyzed, and the relationship between the expression of LRPPRC and Bcl-2 and prognosis was analyzed. Si RNA LRPPRC was used to down-regulate the level of LRPPRC in prostate cancer cells, and the effects on cell proliferation, apoptosis and Bcl-2,Bax,caspase-3 protein expression were observed.
