RNF180启动子区甲基化在前列腺癌中的作用机制

【摘要】:目的通过检测RNF180启动子区甲基化改变来阐明前列腺癌发生和演进过程中RNF180表达的意义。方法分别采用Western blotting、RT-PCR、甲基化特异性PCR(MSP)及亚硫酸氢盐测序(BSP)法检测人前列腺癌细胞系(PC3、LNCap和DU145)与人正常前列腺上皮细胞(RWPE-1)中RNF180的表达情况;采用免疫组化法检测人前列腺癌组织芯片上前列腺癌及癌旁组织中RNF180蛋白的表达。结果前列腺癌细胞中RNF180基因的m RNA及蛋白的表达量明显低于正常前列腺上皮细胞(P0.05),而前列腺癌细胞中RNF180启动子区的甲基化水平却明显高于正常前列腺上皮细胞;RNF180蛋白在前列腺癌组织中的表达量明显低于相应的癌旁组织。结论 RNF180启动子区在前列腺癌细胞中呈高度非完全性甲基化状态,这也可能是RNF180在癌细胞和癌组织中表达下降或沉默的原因之一。
[Abstract]:Objective to investigate the significance of RNF180 expression in the occurrence and evolution of prostate cancer by detecting the methylated changes of RNF180 promoter region. Methods the expression of RNF180 in human prostate cancer cell lines (PC3,LNCap and DU145) and human normal prostate epithelial cells (RWPE-1) was detected by Western blotting,RT-PCR, methylated specific PCR (MSP) and bisulfite sequencing (BSP), respectively. The expression of RNF180 protein in prostate cancer and paracancerous tissues was detected by immunohistochemistry. Results the expression of m RNA and protein of RNF180 gene in prostate cancer cells was significantly lower than that in normal prostate epithelial cells (P 0.05), while the methylated level of RNF180 promoter region in prostate cancer cells was significantly higher than that in normal prostate epithelial cells. The expression of RNF180 protein in prostate cancer tissues was significantly lower than that in corresponding paracancerous tissues. Conclusion the promoter region of RNF180 is highly incomplete methylated in prostate cancer cells, which may be one of the reasons for the decrease or silence of RNF180 expression in cancer cells and cancer tissues.
