高浓度葡萄糖对膀胱癌T24细胞增殖及β-catenin表达的影响

【摘要】:目的探讨高浓度葡萄糖对膀胱癌T24细胞增殖的影响,并探讨其可能的作用机制。方法用含10%FBS的DMEM培养液体外培养膀胱癌T24细胞,将细胞分为5组:空白组(5.5 mmol/L葡萄糖)、对照组(5.5 mmol/L葡萄糖+25 mmol/L甘露醇)、高糖组(10、20、30 mmol/L葡萄糖)。采用MTT法及克隆形成实验检测高浓度葡萄糖对膀胱癌T24细胞增殖的影响;通过实时定量PCR检测膀胱癌T24细胞β-catenin mR NA的表达水平;通过Western blot法检测高浓度葡萄糖对膀胱癌T24细胞β-catenin蛋白表达的影响。结果 MTT法及克隆形成实验结果显示,高浓度葡萄糖显著促进膀胱癌T24细胞的增殖(P0.01);Western blot检测结果显示,高浓度葡萄糖促进T24细胞β-catenin蛋白的表达。实时定量PCR检测结果显示,高浓度葡萄糖促进T24细胞β-catenin mR NA的表达。结论高浓度葡萄糖明显促进膀胱癌T24细胞的增殖,并促进T24细胞β-catenin蛋白和其mRNA的表达,该作用机制可能与β-catenin表达上调有关。

[Abstract]:Objective To study the effect of high-concentration glucose on the proliferation of bladder cancer T24 cells and to explore its possible mechanism of action. Methods T24 cells of bladder cancer were cultured with DMEM containing 10% FBS, and the cells were divided into 5 groups: blank group (5.5 mmol/ L glucose), control group (5.5 mmol/ L glucose + 25 mmol/ L mannitol) and high-sugar group (10,20,30 mmol/ L glucose). The effect of high concentration of glucose on the proliferation of bladder cancer T24 cells was measured by MTT and clone formation. The expression level of T24 cells in bladder cancer T24 cells was detected by real-time quantitative PCR. The effect of high concentration of glucose on the expression of antigen-cata protein in bladder cancer T24 cells was detected by Western blot. Results The results of MTT and clone formation showed that high-concentration glucose significantly promoted the proliferation of T24 cells in bladder cancer (P0.01), and the results of Western blot showed that high-concentration glucose promoted the expression of T-cata protein in T24 cells. The results of real-time quantitative PCR showed that high-concentration glucose promoted the expression of T24 cell-cattenin mRNA. Conclusion High-concentration glucose can promote the proliferation of T24 cells in bladder cancer and promote the expression of T24 cell-cattenin protein and its mRNA. in-house picture: image indicate that high-concentration glucose promote proliferation of T24 cells of bladder cancer: control group; B: blank group; C:10 mmol/ L high-sugar group; D:20 mmol/ L high-sugar group; E:30 mmol/ L high-sugar group
