雄激素受体及共调节因子在雄激素调控勃起信号通路VIP/cAMP中的作用

目的1.探讨雄激素对阴茎海绵体平滑肌细胞(corpus cavernosum smooth muscle cells,CCSMCs)自噬和凋亡的影响,重点研究其对自噬和凋亡桥梁分子BECN1:Bcl-2的调节作用。2.探讨雄激素浓度和雄激素受体(androgen receptor,AR)活性变化对血管活性肠多肽(vasoactive intestinal polypeptide,VIP)调控勃起作用的影响。3.探讨雄激素浓度变化对AR及共调节因子表达的影响,AR及共调节因子信号通路与VIP/c AMP/PKA信号通路的相互作用,进而明确低雄激素浓度时勃起功能维持的机制。方法1.雄性SD大鼠随机分为6组:正常对照组(A组)、手术去势组(B组)、去势后补充十一酸睾酮组(C组)、单独给予AR拮抗剂氟他胺组(D组)、去势后给予氟他胺组(E组)、去势后给予十一酸睾酮和氟他胺组(F组)。2.结合体内电刺激海绵体神经测定海绵体内压力和离体平滑肌条实验测定海绵体平滑肌的舒缩功能,来评估各组大鼠勃起功能情况。3.通过HE染色、Masson三染色、透射电镜观察各组大鼠阴茎结构的变化。4.采用ELISA方法...
