短期PM 2.5 暴露所致肾脏损伤的探讨及复方精油预防作用的研究

目的:随着空气污染的加剧,PM2.5近年来倍受关注,然而研究大多局限于PM2.5对呼吸系统的影响,对肾脏的影响研究较少。本实验探讨了在PM2.5短期暴露造成急性肺损伤的情况下肾脏的损伤情况。同时,探究了复方精油是否可对这种损伤进行干预从而达到预防性治疗的目的。方法:1.PM2.5样品采集及PM2.5悬液制备:本次样本采集于2013年中国河北廊坊,用专门的PM2.5采样器于冬季收集。将收集完成的PM2.5石英滤膜放入超纯水中浸泡、超声处理2-3小时,使PM2.5颗粒与滤膜分离悬浮于超纯水中,后经冷冻干燥去除水分。称取一定量的PM2.5颗粒物,用无菌生理盐水配置成10 mg/m L PM2.5悬液,使用前吹打混匀。2.实验动物处理及实验设计:6-8周的雄性Balb/c小鼠置于适宜的温度(24±1oC),充足的水和食物,规律的昼夜循环环境中。本次的实验分为两部分:(1)第一部分,48只BALB/c小鼠,随机分成两组(n=24),分别为空白对照组和PM2.5暴露实验组。对照组和PM2.5暴露组小鼠分别在实验的第0、2天通过气管灌注方式滴注PM2.5悬液50μL。每次分别在对照组和PM2.5暴露...
