3D打印重建阴道种子细胞制备的实验研究

第一部分体外诱导骨髓间充质干细胞(Bone marrow mesenchymalstem cells,BMSCs)定向分化为内皮细胞、平滑肌细胞、上皮细胞和成纤维细胞目的:探索体外诱导BMSCs向内皮细胞、平滑肌细胞、上皮细胞和成纤维细胞分化的方法,为3D生物打印法重建阴道提供种子细胞。方法:体外培养大鼠BMSCs1 CoCl2模拟乏氧微环境:MTT法检测CoCl2对BMSCs生长的影响,Western blot法检测Co Cl2作用后HIF-1α的表达情况,鉴定CoCl2模拟乏氧微环境的能力。2诱导BMSCs向内皮细胞、平滑肌细胞、上皮细胞和成纤维细胞分化:内皮细胞诱导:CoCl2 100μM预处理BMSCs 24 h体外模拟乏氧微环境,更换为内皮诱导培养基(3%FBS,50 ng/mL VEGF和10 ng/mL bFGF)诱导细胞7/14天。平滑肌细胞诱导:平滑肌细胞诱导培养基(10%FBS,10 ng/mL PDGF-AB和5 ng/mL TGF-β1)诱导BMSCs ...
