细胞角蛋白14基因在非小细胞肺癌中的表达与意义

【摘要】:目的探讨细胞角蛋白14(CK14)基因的表达在非小细胞肺癌(NSCLC)发生、发展及转移中的意义。方法采用免疫组织化学染色法检测110例NSCLC患者癌组织、癌旁组织、正常肺组织中CK14的表达。采用Western blotting法检测12例患者新鲜组织中CK14的表达。应用非条件Logistic回归分析CK14表达水平与NSCLC患者各临床病理参数的关系。结果免疫组织化学染色结果显示,CK14在NSCLC癌组织中高表达,阳性表达率为68.18%,与癌旁组织和正常肺组织相比,差异有统计学意义(P0.001)。CK14表达水平与NSCLC患者性别、组织类型、分化程度、分期以及淋巴结转移情况显著相关(P0.05),与年龄无关(P0.05)。Western blotting结果显示,CK14在NSCLC癌组织中的表达率较癌旁组织和正常肺组织高。非条件Logistic分析显示,低分化癌、病理分期Ⅱ期和Ⅲ期、CK14表达阳性是NSCLC患者发生淋巴结转移的重要危险因素(P0.05)。结论 CK14表达在NSCLC的发生、发展和转移中发挥重要作用。CK14有望成为评估NSCLC生物学行为的重要指标。
[Abstract]:Objective to investigate the expression of cytokeratin 14 (CK14) gene in non-small cell lung cancer (NSCLC). Methods Immunohistochemical staining was used to detect the cancerous and paracancerous tissues in 110 patients with NSCLC. Expression of CK14 in normal lung tissue with Western. Blotting method was used to detect the expression of CK14 in fresh tissues of 12 patients. The expression level of CK14 and the clinicopathological parameters of NSCLC were analyzed by non-conditional Logistic regression analysis. Results Immunohistochemical staining showed that. The expression rate of CK14 was 68.18 in NSCLC tissues, which was higher than that in adjacent tissues and normal lung tissues. There was a significant correlation between the expression of P0.001P. CK14 and sex, tissue type, differentiation, stage and lymph node metastasis of NSCLC patients (P 0.05). The results of Western blotting showed that there was no correlation between age and age. The expression rate of CK14 in NSCLC was higher than that in paracancerous tissues and normal lung tissues.
