中频交变电流联合紫杉醇抗乳腺癌MCF-7细胞作用的研究

【摘要】:目的:研究中频交变电流联合紫杉醇对人乳腺癌MCF-7细胞的作用效果。方法:将对数生长期的乳腺癌MCF-7细胞分别暴露于电刺激、紫杉醇、电刺激与紫杉醇联合作用下,采用cck8法检测各组细胞存活率,观察不同中频交变电流对MCF-7细胞增殖的作用、不同紫杉醇药物浓度影响MCF-7细胞的生长情况、中频交变电流联合紫杉醇对MCF-7细胞增殖的影响、中频交变电流联合紫杉醇作用MCF-7细胞24h后的细胞恢复时间,并运用流式细胞技术分析各组实验作用下对细胞凋亡/死亡、细胞周期的影响。结果:1、随着电流(50—150 mA)的增加,乳腺癌MCF-7细胞的存活率越来越低,且各实验组的存活率与对照组相比,均有显著统计学差异(P0.01)。2、与对照组相比,随着紫杉醇药物浓度(0.01nmol/L—1000nmol/L)的增加,紫杉醇对MCF-7人乳腺癌细胞的抑制率越来越强,各紫杉醇实验组均有显著统计学差异(P0.01),紫杉醇对MCF-7细胞的IC50为31nmol/L。3、中频交变电流(100 kHz,110 mA,30 min)作用MCF-7细胞后,细胞存活率为70±5.24%;联合紫杉醇(IC50)治疗后,细胞存活率为24±1.81%(P0.01)。4、各实验组在24h内治疗后,均可见对MCF-7细胞有不同程度的抑制作用。其中,与对照组相比电刺激组及紫杉醇组的人乳腺癌MCF-7细胞减少了近30%,联合组的人乳腺癌MCF-7细胞减少了近50%,达到半数抑制量(P0.01)。在接下来的48h期间,电刺激组相对于药物组细胞恢复较快;相对于电刺激组,联合组和药物组恢复较慢;最终,联合组细胞存活率最低,仅33.73±4.68%(P0.01)。5、电刺激组及紫杉醇组能够促进MCF-7细胞凋亡,在中频交变电流联合紫杉醇作用后,人乳腺癌MCF-7细胞凋亡率明显增加,与其它三组相比有显著性差异(P0.01)。6、与对照组相比,各实验组G0/G1期细胞明显减少,S期细胞明显增多,其中,联合治疗组G2/M期细胞明显增多(P0.01)。结论:中频交变电流单独使用可以抑制MCF-7细胞增殖,紫杉醇与其联合应用时能表现出协同抗肿瘤细胞作用。
[Abstract]:Objective: to study the effect of intermediate frequency alternating current combined with paclitaxel on human breast cancer MCF-7 cells. Methods: MCF-7 cells in logarithmic phase were exposed to electrical stimulation and paclitaxel respectively. Under the combination of electric stimulation and paclitaxel, the survival rate of MCF-7 cells was detected by cck8 assay, and the effects of different intermediate frequency alternating currents on the proliferation of MCF-7 cells were observed. The growth of MCF-7 cells was affected by different concentrations of paclitaxel, and the proliferation of MCF-7 cells was influenced by intermediate frequency alternating current combined with paclitaxel. The recovery time of MCF-7 cells after 24 hours of treatment with intermediate frequency alternating current combined with paclitaxel was analyzed by flow cytometry. Results: the survival rate of breast cancer MCF-7 cells was lower and lower with the increase of electric current 50-150 Ma, and the survival rate of each experimental group was higher than that of the control group. Compared with the control group, the concentration of paclitaxel increased with 0.01nmol / L ~ (-1) nmol / L). The inhibitory rate of paclitaxel on MCF-7 human breast cancer cells was stronger and stronger, and there was a significant difference among each paclitaxel experimental group (P0.01). The IC50 of paclitaxel on MCF-7 cells was 31nmol / L ~ (3), and the medium frequency alternating current was 100kHz ~ 110mA ~ (-1) for 30 min). The cell survival rate was 70 卤5.24; The cell survival rate was 24 卤1.81% after treatment with paclitaxel IC50, and the survival rate of each experimental group was 24 卤1.81% within 24 hours. Compared with the control group, the number of MCF-7 cells in the electrical stimulation group and paclitaxel group decreased by nearly 30%. In the combined group, the MCF-7 cells of breast cancer decreased by nearly 50%, reaching the half inhibitory amount (P 0.01). During the next 48 hours, the cells recovered faster in the electric stimulation group than in the drug group. Compared with the electrical stimulation group, the recovery was slower in the combined group and the drug group. In the end, the survival rate of the combined group was the lowest, only 33.73 卤4.68% (P 0.01). The electric stimulation group and paclitaxel group could promote the apoptosis of MCF-7 cells. The apoptosis rate of human breast cancer MCF-7 cells increased significantly after the treatment of intermediate frequency alternating current combined with paclitaxel, which was significantly different from the other three groups (P 0.01) and compared with the control group (P < 0.05). The number of G _ 0 / G _ 1 phase cells in each experimental group was significantly decreased, and the number of S phase cells was significantly increased. In the combined treatment group, the number of G2 / M phase cells increased significantly (P 0.01). Conclusion: the intermediate frequency alternating current alone can inhibit the proliferation of MCF-7 cells. Paclitaxel combined with paclitaxel showed synergistic anti-tumor effect.
