扶正康复合剂与顺铂联用抑制移植性肺癌的实验研究

【摘要】:目的:研究扶正康复合剂对移植性肺癌的抑制作用,并探讨其作用机制。方法:将48只NOD/SCID裸鼠随机分为8组:模型对照组,顺铂组,扶正康复合剂低、中、高剂量组,顺铂+扶正康复合剂低、中、高剂量组,共8组,每组6只,皮下接种H1975细胞建立移植性裸鼠肺癌模型,接种15 d后开始给药,连续给药7 d,称取体重及瘤重,计算抑瘤率,ELISA法检测裸鼠血清BUN、Cr、TNF-α、IFN-γ、IL-2、IL-6的表达水平,免疫组化法检测瘤体组织VEGF、Bcl-2、Bax、Survivin的表达情况,q-PCR法检测瘤体组织VEGF、Bcl-2、Bax、Survivin mRNA的表达水平。结果:扶正康复合剂高剂量组、顺铂组、顺铂+扶正康复合剂低、中、高剂量组与模型对照组相比:荷瘤裸鼠瘤质量显著减轻(P0.05),血清TNF-α水平显著升高(P0.05),IL-2水平显著升高(P0.05),血清IL-6水平显著降低(P0.05),Bcl-2表达量显著减少,Bax表达量增加。其中顺铂+扶正康复合剂高剂量给药组裸鼠抑瘤率最高,两者具有协同作用。与顺铂组相比:顺铂+扶正康复合剂高剂量组裸鼠血清Cr、BUN水平显著降低(P0.01),顺铂+扶正康复合剂中、高剂量组裸鼠血清TNF-α、INF-γ水平显著升高(P0.01);顺铂+扶正康复合剂高剂量组Bcl-2、Survivin、VEGF表达量降低,Bax表达量增加。结论:扶正康复合剂与顺铂联用能够降低顺铂的肾损伤作用,调节机体的Th1/Th2细胞免疫平衡,通过抑制Bcl-2、Survivin、VEGF并促进Bax的表达,进而促进肺癌细胞的凋亡。
[Abstract]:Objective: to study the inhibitory effect of Fuzhenghuangji on transplanted lung cancer and its mechanism. Methods: 48 NOD/SCID nude mice were randomly divided into 8 groups: model control group and cisplatin group. The model of transplanted nude mice lung cancer was established by inoculating H1975 cells subcutaneously into 8 groups of low, middle and high dosage groups of low, medium and high dose of Fuzheng Kangfeng mixture and low, middle and high dose groups of Cisplatin Fuzheng Kangfeng mixture. After 15 days of inoculation, the drug was administered continuously for 7 days. The weight and tumor weight were weighed. The inhibition rate of tumor was calculated and Elisa was used to detect IL-2 in nude mice serum BUNCU TNF- 伪 and IFN- 纬. The expression of IL-6 was detected by immunohistochemical method. The expression of survivin was detected by immunohistochemical method. VEGF was detected by q-PCR. Results: high dose group, cisplatin group, cisplatin group, cisplatin Fuzheng convalescent mixture were low and medium. Compared with the model control group, the tumor quality of the high dose group decreased significantly (P 0.05), and the serum TNF- 伪 level increased significantly (P 0.05). The level of IL-2 was significantly higher than that of P0.05, and the level of serum IL-6 was significantly lower than that of P0.05, and the expression of Bcl-2 was significantly decreased. The expression of Bax was increased. The tumor inhibition rate of nude mice was the highest in the high dose group. Compared with the cisplatin group, the serum Cr of the nude mice in the high dose group was higher than that in the cisplatin Fuzheng Kang mixture group. The level of BUN decreased significantly (P 0.01). In Cisplatin Fuzheng Rehabilitation mixture, the serum TNF- 伪 / INF- 纬 level in high dose group increased significantly (P 0.01). The expression of VEGF was decreased in the high dose group of cisplatin Fuzheng convalescent mixture. Conclusion: the combination of Fuzhenghuangji and cisplatin can reduce the renal injury of cisplatin and regulate the immune balance of Th1/Th2 cells by inhibiting Bcl-2. Survivin VEGF also promotes the expression of Bax and promotes the apoptosis of lung cancer cells.
