HER2和TSPAN1蛋白在胃癌组织中的表达及临床病理意义

【摘要】:目的:(1)探讨HER2和TSPAN1蛋白在胃癌发生发展中的作用及意义。(2)探讨HER2和TSPAN1蛋白的表达与胃癌患者年龄、性别、肿瘤大小、部位、分化程度、转移、Lauren分型和TNM分期的关系及意义。(3)探讨HER2和TSPAN1蛋白在胃癌发展中是否有协同关系。方法:采用Envision免疫组织化学法(二步法)对64例胃癌组织和癌旁粘膜组织进行HER2和TSPAN1蛋白表达的检测,同时选取20例正常胃粘膜组织作为对照。结果:(1)64例胃癌组织中HER2蛋白阳性表达率23.4%(15/64)显著高于20例正常胃粘膜组织0%(0/20),经统计学分析,差异具有显著性(P0.05);64例癌旁粘膜组织中HER2蛋白阳性表达率10.9%(7/64)高于20例正常胃粘膜组织0%(0/20),经统计学分析,差异无显著性(P0.05);64例胃癌组织中HER2蛋白阳性表达率23.4%(15/64)高于64例癌旁粘膜组织10.9%(7/64),经统计学分析,差异无显著性(P0.05)。(2)Lauren分型42例肠型胃癌组织中HER2蛋白阳性表达率33.3%(14/42)显著高于22例弥漫型及混合型胃癌组织4.5%(1/22),经统计学分析,差异具有显著性(P0.05)。13例高分化胃癌组织38.5%(5/13)、29例中分化胃癌组织中HER2蛋白阳性表达率31.0%(9/29)显著高于22例低分化胃癌组织4.5%(1/22),经统计学分析,差异具有显著性(P0.05)。38例淋巴结转移胃癌组织中HER2蛋白阳性表达率36.8%(14/38)显著高于26例无淋巴结转移胃癌组织3.8%(1/26),经统计学分析,差异具有显著性(P0.05)。HER2蛋白阳性表达与患者年龄、性别、肿瘤大小、部位、TNM分期无关(P0.05)。(3)64例胃癌组织中TSPAN1蛋白阳性表达率68.8%(44/64)显著高于20例正常胃粘膜组织0%(0/20),经统计学分析,差异具有显著性(P0.05);64例癌旁粘膜组织中TSPAN1蛋白阳性表达率23.4%(15/64)显著高于20例正常胃粘膜组织0%(0/20),经统计学分析,差异有显著性(P0.05);64例胃癌组织中TSPAN1蛋白阳性表达率68.8%(44/64)显著高于64例癌旁粘膜组织23.4%(15/64),经统计学分析,差异具有显著性(P0.05)。(4)Lauren分型42例肠型胃癌组织中TSPAN1蛋白阳性表达率52.4%(22/42)显著低于22例弥漫型及混合型胃癌组织100%(22/22),经统计学分析,差异具有显著性(P0.05)。22例低分化胃癌组织100%(22/22)、29例中分化胃癌组织中TSPAN1蛋白阳性表达率62.1%(18/29)显著高于13例高分化胃癌组织30.8%(4/13),经统计学分析,差异具有显著性(P0.05)。TSPAN1蛋白阳性表达与患者年龄、性别、肿瘤大小、部位、转移、TNM分期无关。(5)胃癌组织中HER2和TSPAN1蛋白表达有一定的相关性(X2=7.107,r=0.349,P=0.003)。结论:(1)HER2和TSPAN1蛋白阳性表达与胃癌的发生发展有关。(2)HER2和TSPAN1蛋白阳性表达与胃癌分化程度及Lauren分型有关,而且HER2蛋白阳性表达与胃癌患者的转移也有关。(3)HER2和TSPAN1蛋白阳性表达与胃癌患者年龄、性别、肿瘤大小、部位、TNM分期无关,TSPAN1蛋白阳性表达与转移无关。
[Abstract]:Objective to explore the role and significance of HER2 and TSPAN1 proteins in the development of gastric cancer. (2) to explore the relationship between the expression of HER2 and TSPAN1 proteins and the age, sex, tumor size, location and differentiation of gastric cancer patients. The relationship between TNM staging and HER2 classification) to explore whether there is a synergistic relationship between HER2 and TSPAN1 protein in the development of gastric cancer. Methods: 64 gastric cancer tissues and adjacent mucosa tissues were studied by Envision immunohistochemical method. HER2 and TSPAN1 protein expression were detected. At the same time, 20 normal gastric mucosa tissues were selected as control. Results the positive expression rate of HER2 protein in 64 cases of gastric cancer was significantly higher than that of 20 cases of normal gastric mucosa tissue, and the positive rate of HER2 protein in 64 cases of gastric cancer was significantly higher than that of 20 cases of normal gastric mucosa tissue. The positive expression rate of HER2 protein in 64 cases of paracancerous mucosa was significantly higher than that of 20 cases of normal gastric mucosa (10.9% / 64), and was significantly higher than that of 20 cases of normal gastric mucosa tissue (0 / 20%). The positive expression rate of HER2 protein in 64 cases of gastric cancer was 23.4% (15 / 64) higher than that of 10.9% (10.9%) in 64 cases of gastric cancer. The positive expression rate of HER2 protein in 42 cases of intestinal type gastric carcinoma was significantly higher than that of 22 cases of diffuse type and mixed type of gastric cancer. The positive expression rate of HER2 protein in 42 cases of intestinal type gastric carcinoma was significantly higher than that of 22 cases of diffuse type and mixed type of gastric carcinoma. The positive expression rate of HER2 protein in 29 cases of well-differentiated gastric carcinoma was significantly higher than that in 22 cases of poorly differentiated gastric carcinoma (P 0.05). The expression rate of HER2 protein in 29 cases of well-differentiated gastric carcinoma was significantly higher than that of 22 cases of poorly differentiated gastric carcinoma, and the expression rate of HER2 protein was significantly higher than that in 22 cases of poorly differentiated gastric carcinoma, and the expression rate of HER2 protein in 29 cases was significantly higher than that in 22 cases of poorly differentiated gastric carcinoma. The positive expression rate of HER2 protein in 38 cases with lymph node metastasis was significantly higher than that in 26 cases without lymph node metastasis. The positive expression rate of HER2 protein in 38 cases of lymph node metastasis was significantly higher than that of 26 cases without lymph node metastasis. By statistical analysis, the positive expression of HER2 protein in patients with lymph node metastasis was significantly higher than that in patients with age and sex. The positive expression rate of TSPAN1 protein in 64 cases of gastric cancer was significantly higher than that in 20 cases of normal gastric mucosa. The positive expression rate of TSPAN1 protein in 64 paracancerous mucosa tissues was significantly higher than that in 20 normal gastric mucosa tissues. The positive expression rate of TSPAN1 protein in 64 cases of gastric cancer was significantly higher than that in the adjacent mucosa of 64 cases (68.8% / 64%), which was significantly higher than that in the adjacent mucosa tissue of 64 cases (23.4% / 64%). The positive expression rate of TSPAN1 protein in 42 cases of intestinal type gastric carcinoma was significantly lower than that in 22 cases of diffuse type and mixed type of gastric cancer. The expression rate of TSPAN1 protein in 42 cases of intestinal type gastric carcinoma was significantly lower than that of 22 cases of diffuse type and mixed type of gastric carcinoma. The positive expression rate of TSPAN1 protein in 22 cases of poorly differentiated gastric carcinoma was significantly higher than that in 13 cases of well-differentiated gastric carcinoma. The positive expression rate of TSPAN1 protein was significantly higher than that in 13 cases of well-differentiated gastric carcinoma. The difference was statistically significant between P0.05. TSPAN1 protein expression and age. Sex, tumor size, location, There was a correlation between the expression of HER2 and TSPAN1 protein in gastric cancer tissues. Conclusion the positive expression of HER2 and TSPAN1 proteins is related to the occurrence and development of gastric carcinoma, and the expression of HER2 and TSPAN1 protein is related to the differentiation of gastric carcinoma and the type of Lauren. Moreover, the positive expression of HER2 protein was related to metastasis of gastric cancer patients. The positive expression of HER2 and TSPAN1 protein was not related to age, sex, tumor size and tumor stage of gastric cancer patients. There was no correlation between the positive expression of TSPAN1 protein and metastasis.
