突变型P53和CyclinD1蛋白在宫颈癌组织中表达的意义

【摘要】：目的:通过研究突变型P53蛋白、Cyclin D1蛋白与宫颈癌组织学分级、病理类型、淋巴结转移等病理特征之间相关性以及二者自身之间的关联,探讨二者在宫颈癌发生发展过程中所具有的意义。方法:应用免疫组化PV-6000二步法对2012-2015年青岛大学附属医院18例正常宫颈组织、10例低级别宫颈上皮内瘤变组织(CINI级)、29例高级别宫颈上皮内瘤变组织(CINII-III级)、42例宫颈癌组织进行免疫组织化学染色,检测各组突变型P53及Cyclin D1蛋白表达情况。结果:(1)突变型P53蛋白在正常宫颈组、CINI组、CINII-III组、宫颈癌组中阳性表达率分别为0.00%、10.00%、44.82%、66.71%,呈明显递增趋势,差异有统计学意义(x2=27.763,P0.05)。Cyclin D1蛋白在正常宫颈组、CINI组、CINII-III组、宫颈癌组的阳性表达率分别为16.67%、20.00%、58.62%、83.33%,且差异有统计学意义(x2=34.270,P0.05)。(2)突变型P53和Cyclin D1蛋白在宫颈癌组织中的表达状况均与组织分化程度、淋巴结有无转移相关(P0.05),与病理类型、临床分期无相关性(P0.05)。(3)在宫颈癌组织中突变型P53蛋白、Cyclin D1蛋白的阳性表达呈正相关(r=0.305,P0.05)。结论:突变型P53蛋白、Cyclin D1蛋白参与宫颈病变进展,并与癌细胞分化程度及恶性程度有一定的关系,二者呈协同作用,共同促进宫颈病变的进展。
[Abstract]:Objective: to study the correlation between the mutant p53 protein Cyclin D1 protein and the histological grade, pathological type, lymph node metastasis and other pathological features of cervical cancer. To explore the significance of the two methods in the occurrence and development of cervical cancer. Methods: immunohistochemical PV-6000 two-step method was used to study 18 cases of normal cervix tissues in Qingdao University affiliated Hospital from 2012-2015 to 10 cases of low grade cervical intraepithelial neoplasia (CINI). 29 cases of high grade cervical intraepithelial neoplasia and 42 cases of cervical carcinoma of grade CINII-III were stained with immunohistochemical staining. Results the positive expression rates of p53 and Cyclin D1 protein in normal cervix cervix group and cervical carcinoma group were 0.0010.00 and 44.82%, respectively. There was significant difference in the expression of p53 and Cyclin D1 protein in normal cervix carcinoma group (CINI group) and cervical carcinoma group (P < 0.01). The positive expression rates of p53 and Cyclin D1 protein in cervical carcinoma group were 16.6767 ~ 20.00% and 88.62.33%, respectively, and the difference was statistically significant (P < 0.01) the expression of p53 and Cyclin D1 protein in cervical carcinoma tissues were all related to the degree of tissue differentiation. There was a positive correlation between the positive expression of mutant p53 protein and Cyclin D1 protein in cervical cancer tissues. Conclusion: the mutant p53 protein Cyclin D1 protein is involved in the progression of cervical lesions. And there is a certain relationship with the degree of differentiation and malignancy of cancer cells, both of which have synergistic effect to promote the progress of cervical lesions.
