胃癌EMT信号通路相关长链非编码RNA的表达及临床意义

【摘要】:目的:研究上皮-间质转化(Epithelial-mesenchymal transition,EMT)信号通路相关长链非编码RNA(Long non-coding RNA,LncRNA)在胃癌组织及癌旁胃粘膜组织中的差异表达,探索代表性EMT相关lncRNA在胃癌临床诊断中的潜在价值。方法:收集胃癌组织及癌旁胃粘膜组织,应用LncPath Human EMT Pathway LncRNA Microarray检测胃癌组织和癌旁胃粘膜组织EMT相关lncRNA和相关靶基因的表达水平,筛选出表达差异明显的lncRNA,并绘制与相关靶基因相互作用图。实时定量逆转录-聚合酶链式反应(quantitative reverse transcription-polymerase chain reaction,qRT-PCR)检测胃癌组织、癌旁胃粘膜组织、胃癌细胞株BGC-823及正常胃粘膜细胞GES-1 lncRNA metallothionein 1J,pseudogene(MT1JP)和mRNA-metallothionein 1M(MT1M)的表达水平。结合临床资料分析,MT1JP和MT1M表达水平与患者临床病理因素之间的相关性,探索其在临床诊断中的潜在价值。结果:胃癌组织与癌旁胃粘膜组织之间表达差异超过2倍lncRNA有26条,有16条明显上调,10条明显下调;qRT-PCR检测结果LncRNA-MT1JP和靶基因mRNAMT1M的表达水平在胃癌组织中降低,MT1JP表达水平与肿瘤大小、分期、淋巴转移、远处转移、TNM分期相关(P0.05)。MT1JP在胃癌前病变、早期胃癌中及进展期胃癌中表达依次降低。结论:胃癌中大量EMT相关lncRNA异常表达,可能通过EMT相关信号通路在胃癌发展中发挥调节作用。MT1JP和MT1M在胃癌组织中低表达,与肿瘤的分期及浸润程度具有相关性。MT1JP在癌前病变及早期胃癌中低表达,可能是潜在的胃癌分子诊断和治疗的靶标。
[Abstract]:Objective: to investigate the differential expression of Epithelial-mesenchymal transition (EMT) signaling pathway associated with long chain noncoding RNA(Long non-coding RNAs in gastric carcinoma and adjacent gastric mucosa. To explore the potential value of representative EMT associated lncRNA in the clinical diagnosis of gastric cancer methods: gastric cancer tissues and adjacent gastric mucosa tissues were collected. LncPath Human EMT Pathway LncRNA Microarray was used to detect the expression of EMT related lncRNA and related target genes in gastric carcinoma and adjacent gastric mucosa. The differentially expressed LNC RNAs were screened, and the interaction diagrams with related target genes were plotted. Real-time quantitative reverse transcription-polymerase chain reactionsqRT-PCRs were used to detect gastric cancer tissues and adjacent gastric mucosa. The expression levels of GES-1 lncRNA metallothionein 1J BGC-823 and mRNA-metallothionein 1Mt1MP) in gastric cancer cell line BGC-823 and normal gastric mucosal cells were analyzed. The correlation between the expression levels of MT1M and clinicopathological factors was analyzed in combination with clinical data. Results: the difference of expression between gastric cancer tissue and adjacent gastric mucosa was more than 2 times lncRNA. The expression level of LncRNA-MT1JP and target gene mRNAMT1M in gastric cancer tissues decreased significantly. The expression level of MT1JP and tumor size, staging, lymphatic metastasis and distant metastasis were correlated with P0.05. MT1JP in gastric precancerous lesions. Conclusion: a large number of abnormal expression of EMT related lncRNA may play a regulatory role in the development of gastric cancer through EMT related signaling pathway. MT1 JP and MT1M may play a low expression in gastric cancer tissues. The low expression of MT1JP in precancerous lesions and early gastric cancer may be a potential target for molecular diagnosis and treatment of gastric cancer.
