喉鳞状细胞癌中ALDOA、G6PD和PFK1的表达及临床意义

【摘要】:目的:研究喉鳞状细胞癌肿瘤组织中醛缩酶(aldolase A,ALDOA)、葡萄糖6磷酸脱氢酶(Glucose-6-phosphate,G6PD)、磷酸果糖激酶1(Phosphofructokinase-1,PFK1)的表达情况、以及三者间关系及与临床病理参数的关系,进一步探讨肿瘤组织细胞中糖酵解和磷酸戊糖途径在代谢等方面的潜在关系,并揭示这三种蛋白与喉鳞状细胞癌的发生、增殖、侵袭、转移等过程中的作用。方法:选取在河北医科大学第二医院2015年6月-2016年12月间40例行手术治疗的喉癌病人的标本,并经病理鉴定,确定这40例喉肿瘤标本均为鳞状细胞癌,此外,选取40例癌旁组织(距肿瘤边沿距离0.5cm,经病理证实未见肿瘤细胞浸润)作为对照,喉鳞状细胞癌组和对照组患者手术前均未接受过放化疗及其他相关治疗。本实验采取免疫组织化学染色法中免疫酶细胞化学技术检测喉鳞状细胞癌肿瘤组织及癌旁组织中ALDOA、PFK1和G6PD蛋白的表达情况,实验数据通过SPSS13.0软件分析,采用χ2检验方法分析喉癌及癌旁组织中这三种蛋白表达的阳性率,并分析这三者水平之间的差异,还分别分析这三种蛋白与喉癌患者的临床上的病理参数的关系,采用Spearman等级相关方法检测ALDOA、G6PD和PFK1蛋白间的相互关系,统计学以双侧P0.05有显著意义。结果:1在喉癌和癌旁组织中ALDOA、G6PD、PFK1蛋白表达的阳性率分别为65.0%和15.0%、55.0%和20.0%、52.5%和30.0%,各两组间相比,差异均有显著的统计学意义(P0.05);在喉癌组中ALDOA、G6PD、PFK1蛋白在临床分期为I~II期及III~IV期表达的阳性率分别为44.4%和81.8%、33.3%和72.7%、33.3%和68.2%,各两组间差异比较均有显著的统计学意义(P0.05);在病理为高分化组及中~低分化组的喉癌患者中ALDOA、G6PD、PFK1蛋白表达的阳性率分别为47.6%和84.2%、33.3%和78.9%、28.6%和78.9%,各两组间差异对比均有显著的统计学意义;ALDOA、G6PD、PFK1蛋白分别在有和无淋巴结转移组的喉癌患者中表达的阳性分别为78.6%和33.3%、67.9%和25.0%、64.3%和25.0%,各两组间相比差异均有显著统计学意义(P0.05);在年龄≥60岁组及60岁组的喉癌组中ALDOA、G6PD、PFK1蛋白表达的阳性率分别为65.2%和65.7%、47.8%和64.7%、47.8%和58.8%,各两组间差异比较均无显著统计学意义(P0.05);ALDOA、G6PD、PFK1蛋白在吸烟≥20支/天组及20支/天组的喉癌病人中表达的阳性率分别为61.5%和66.7%、38.5%和63.0%、38.5%和59.3%,各两组间结果比较均无显著统计学意义(P0.05)。2在喉癌的病人中ALDOA蛋白与G6PD蛋白的水平呈正相关(相关系数r=0.601,P0.05);而且ALDOA蛋白与PFK1蛋白在喉癌中的水平也呈正相关(相关系数r=0.457,P0.05);还有PFK1蛋白与G6PD蛋白的水平也呈正相关表现(相关系数r=0.750,P0.05)。结论:1在喉鳞状细胞癌组中ALDOA、G6PD及PFK1蛋白水平均明显高于癌旁组,而且这三种蛋白的水平与淋巴结转移与否、病理分化程度有关,另外三者的水平均还与喉癌临床分期相关,但是这三种蛋白的水平都与病人的年龄以及吸烟的数量无关。由此可见这三种蛋白在喉鳞状细胞癌的形成、生长、细胞的增殖及肿瘤的转移中起着一定的作用。2喉癌组中PFK1蛋白、G6PD蛋白水平均与ALDOA蛋白水平相关,ALDOA可能是联系喉癌中糖酵解和磷酸戊糖途径的一个重要因子,这三种蛋白在肿瘤的形成、生长和转移浸润等过程中可能相互促进,紧密联系,有协同作用。
[Abstract]:Objective: To study the tumor aldolase in laryngeal squamous cell carcinoma (aldolase, A, ALDOA), glucose 6 phosphate dehydrogenase (Glucose-6-phosphate, G6PD), phosphofructokinase 1 (Phosphofructokinase-1, PFK1) expression, and the relationship between the three and their relationship with clinical pathological parameters, to further explore the potential relationship between tumor cell glycolysis and the pentose phosphate pathway in the metabolism and other aspects, and reveals the three proteins and laryngeal squamous cell carcinoma, proliferation, invasion and metastasis process. Methods: a total of Second Hospital of Hebei Medical University in June 2015 -2016 year in December 40 cases of surgical treatment of patients with laryngeal cancer specimens by pathology and identification, to determine which 40 cases of laryngeal tumor specimens were squamous cell carcinoma, in addition, a total of 40 cases of paracancerous tissues (from tumor edge distance 0.5cm, confirmed by pathology and no tumor cell invasion) as control, laryngeal squamous Cell carcinoma group and control group patients before surgery had not received chemotherapy and other related treatment. This experiment adopt immunohistochemical staining ALDOA immunocytochemistry technique in detection of laryngeal squamous cell carcinoma tissues and adjacent tissues, the expression of PFK1 and G6PD proteins, the analysis of experimental data by using SPSS13.0 software. 2 test method for the analysis of the positive expression rate of the three proteins in laryngeal carcinoma and adjacent tissues, and analyze the difference between the three levels of relationship were also pathological analysis of the three proteins and clinical parameters of patients with laryngeal carcinoma on, using Spearman rank correlation method to detect ALDOA, G6PD and PFK1 protein interaction the statistics on bilateral P0.05 was significant. Results: ALDOA 1 in laryngeal carcinoma and adjacent tissues of G6PD, the positive rate of PFK1 protein expression were 65% and 15%, 55% and 20%, 52.5% and 30%, two in each group Compared with statistical significant differences (P0.05); ALDOA, G6PD in laryngeal carcinoma group, PFK1 protein in clinical stage positive expression rate of I~II and III~IV were 44.4% and 81.8%, 33.3% and 72.7%, 33.3% and 68.2%, the difference between the two groups were significant (P0.05) in high differentiation group; pathology and in low differentiation group of laryngeal cancer patients in ALDOA, G6PD, the positive rate of PFK1 protein expression were 47.6% and 84.2%, 33.3% and 78.9%, 28.6% and 78.9%, the difference between the two groups comparison was statistically significant; ALDOA, G6PD, PFK1 protein respectively with and without the expression of lymph node metastasis in patients with laryngeal cancer were respectively 78.6% and 33.3%, 67.9% and 25%, 64.3% and 25%, there was significant difference compared between the two groups (P0.05); ALDOA in aged 60 and 60 years of age group G6PD laryngeal carcinoma group, the positive rate of PFK1 protein expression Were 65.2% and 65.7%, 47.8% and 64.7%, 47.8% and 58.8%, the difference between the two groups were not statistically significant (P0.05); ALDOA, G6PD, PFK1 protein in smoking more than 20 cigarettes / day group and 20 day group / positive expression rate of laryngeal cancer patients were 61.5% and 66.7%, 38.5% and 63%, 38.5% and 59.3%, the two groups showed no statistically significant results (P0.05) levels were positively correlated with ALDOA in laryngeal carcinoma.2 protein and G6PD protein (correlation coefficient r=0.601, P0.05); and the level of ALDOA protein and PFK1 protein in laryngeal carcinoma and Cheng Zhengxiang (correlation coefficient r=0.457, P0.05); and PFK1 protein and G6PD protein expression level was positively correlated (correlation coefficient r=0.750, P0.05). Conclusion: 1 in laryngeal squamous cell carcinoma group ALDOA, G6PD and PFK1 protein levels were significantly higher than those in adjacent groups, and the water level of the three proteins with lymph node metastasis and pathological differentiation. To certain extent, the other three levels were also related and clinical stage, but these three kinds of protein levels and patient's age and smoking. Thus independent of the number of formation of these three proteins in laryngeal squamous cell carcinoma growth, plays a role of PFK1 protein in laryngeal carcinoma.2 group the proliferation and metastasis of tumor cells in the G6PD protein levels were correlated with ALDOA protein levels, ALDOA may be an important factor in the relationship between laryngeal glycolysis and the pentose phosphate pathway, the formation of these three proteins in tumor growth and metastasis, infiltration can promote each other, closely, have synergistic effect.
