SFRP1基因甲基化及其蛋白在肺腺癌中的表达及临床意义

【摘要】:目的探讨SFRP1基因甲基化及其蛋白在肺腺癌组织中的表达。方法收集手术切除的肺腺癌标本60例和癌旁组织30例,采用甲基化特异性PCR法、实时荧光定量PCR法和免疫组织化学染色法分别检测SFPR1基因甲基化、m RNA及蛋白的表达情况。结果 SFRP1基因甲基化率在肺腺癌中(58.3%)表达明显高于癌旁组织(20.0%)(P=0.001)。SFRP1 m RNA在肺腺癌(0.847±0.297)中的表达明显低于癌旁组织(2.019±0.445)(P0.01)。SFRP1蛋白在肺腺癌中的阳性表达率为38.3%(23/60),明显低于癌旁组织中的表达率83.3%(25/30)(P0.001);SFRP1蛋白表达与肺腺癌组织分化程度、临床分期及淋巴结转移情况有关(均P0.05)。SFRP1基因甲基化与其蛋白表达缺失存在相关性(r=-0.585,P0.001)。结论 SFRP1蛋白的表达降低可能是由于SFRP1基因甲基化所致,SFRP1基因甲基化及其蛋白表达与肺腺癌组织分化程度及淋巴结转移情况密切相关。
[Abstract]:Objective to investigate the expression of SFRP1 gene methylation and its protein in lung adenocarcinoma. Methods 60 cases of lung adenocarcinoma and 30 cases of adjacent tissues were collected by methylation specific PCR. The expression of methylated m RNA and protein of SFPR1 gene was detected by real-time fluorescence quantitative PCR and immunohistochemical staining. Results the methylation rate of SFRP1 gene in lung adenocarcinoma was significantly higher than that in paracancerous tissue (P 0.001). SFRP1 m RNA was higher in lung adenocarcinoma than that in paracancerous tissue. The expression of SFRP1 protein in adenocarcinoma was significantly lower than that in paracancerous tissue (2.019 卤0.445), and the positive rate of SFRP1 protein in lung adenocarcinoma was 38.3%. It was significantly lower than that in paracancerous tissue (83.3% 30%) and the degree of differentiation of lung adenocarcinoma, and the expression of SFRP1 protein was significantly lower than that in paracancerous tissue (P 0.001). Clinical stage and lymph node metastasis were related to the methylation of SFRP1 gene and its protein deletion. Conclusion the decreased expression of SFRP1 protein may be due to the methylation of SFRP1 gene and its protein. The expression was closely related to the degree of differentiation and lymph node metastasis of lung adenocarcinoma.
