SBP1在口腔肿瘤中表达的研究

【摘要】:目的硒结合蛋白1(Selenium binding protein 1,SBP1)位于染色体lq21-22上,是一种特殊的含硒蛋白质,主要是通过促进肿瘤细胞的凋亡,以抑制肿瘤的发生。本课题检测SBP1在涎腺肿瘤(salivary gland tumor)中的表达水平,并明确其与口腔肿瘤细胞—SCC3细胞的发生发展、侵袭以及转移的关系,探讨SBP1作为口腔肿瘤的肿瘤生物标志物,评估临床预后的潜能。方法收集手术切除的经病理证实的15例良性涎腺肿瘤及瘤旁组织(多形性腺瘤),分别提取组织RNA,采用实时荧光定量PCR检测组织中SBP1的表达水平。分别构建慢病毒载体介导的SBP1的沉默和过表达的SCC3细胞,实时定量PCR检测慢病毒转染效率。应用细胞划痕实验检测SBP1沉默和过表达后,肿瘤细胞迁移能力的改变;采用MTT检测SBP1沉默和过表达后,肿瘤细胞增殖能力的变化。结果1.慢病毒能够有效改变目的基因表达,具有很高的转染效率。2.实时荧光定量PCR检测SBP1在涎腺肿瘤组织中的表达明显低于正常涎腺组织(P0.05)。3.划痕实验检测沉默SBP1能使SCC3细胞的迁移能力加快,SBP1过表达时SCC3细胞的迁移能力减慢。4.MTT实验显示沉默SBP1促进SCC3细胞的增殖,过表达SBP1抑制SCC3细胞的增殖。结论1.SBP1在涎腺肿瘤组织和瘤旁组织的表达存在差异性,结果具有统计学意义。2.SBP1具有抑制SCC3细胞的增殖与迁移能力。3.SBP1可能是口腔肿瘤的早期诊断指标之一。
[Abstract]:Objective selenium binding protein 1 SBP1) is a special protein containing selenium, which is located on chromosome lq21-22, mainly by promoting apoptosis of tumor cells to inhibit the occurrence of tumor. In this study, the expression level of SBP1 in salivary gland tumor was detected. The relationship between SBP1 and the occurrence, development, invasion and metastasis of oral tumor cell line-SCC3 was also discussed, and SBP1 was used as a biomarker of oral tumor. Methods Fifteen patients with pathologically proved benign salivary gland tumors and adjacent tissues (pleomorphic adenoma) were collected and their SBP1 expression was detected by real-time fluorescence quantitative PCR. Lentivirus vector mediated SBP1 silencing and overexpression of SCC3 cells were constructed. Real-time quantitative PCR was used to detect the efficiency of lentivirus transfection. Cell scratch assay was used to detect the change of tumor cell migration ability after SBP1 silencing and overexpression, and MTT was used to detect SBP1 silencing and overexpression. Results 1. Lentivirus can effectively change the expression of target gene. 2. The expression of SBP1 in salivary gland tumor tissue was significantly lower than that in normal salivary gland tissue (P 0.05N. 3). SBP1 silencing by scratch test could accelerate the migration ability of SCC3 cells. 4. MTT assay showed that silencing SBP1 could promote the proliferation of SCC3 cells. Overexpression of SBP1 inhibits the proliferation of SCC3 cells. 1. There are differences in the expression of SBP1 in salivary gland tumor tissues and adjacent tissues. Results SBP1 has the ability to inhibit the proliferation and migration of SCC3 cells. 3. SBP1 may be one of the early diagnostic markers of oral tumor.
