马来酸噻吗洛尔对血管瘤动物模型鸡冠血管损伤效应

【摘要】:目的研究毛细血管瘤模型鸡冠经马来酸噻吗洛尔滴眼液处理后组织病理学变化以及血管内皮细胞凋亡情况,初步探讨马来酸噻吗洛尔局部湿敷对毛细血管的损伤效应及联合聚桂醇注射治疗提高聚桂醇治疗效果的可行性。方法取6月龄健康雄性鸡30只,随机分为五组,每组6只.对照组E(0.9%的氯化钠溶液外用)。实验组:A组用马来酸噻吗洛尔滴眼液外用;B组用醋酸曲安奈德注射液外用;C组予以聚桂醇注射液注射治疗;D组马来酸噻吗洛尔滴眼液与聚桂醇注射液联合应用。观察各组经药物处理后动物模型的一般情况,分别于处理后第7天,14天,21天,28天从ABCDE五个组随机选取一只鸡,取材后,10%福尔马林溶液固定,常规HE染色,在光学显微镜下选5个高倍镜视野(10X40倍)并拍摄照片,统计毛细血管数目,计算其平均值,并行TUNEL染色观察毛细血管内皮细胞凋亡情况。结果实验组鸡冠与对照组相比,其颜色均有不同程度变浅变淡;HE染色切片光镜下观察可见毛细血管数目减少,与E组相比,差异均有统计学意义(p均0.05);行TUNEL染色观察可见噻吗洛尔干预组真皮乳头层凋亡毛细血管内皮细胞,聚桂醇干预组真皮网状层可见少量凋亡毛细血管内皮细胞,并且随着观察周期的延长,凋亡细胞数目逐渐增多,E组与A,B,C,D组相比较,毛细血管凋亡数目差异均有统计学意义(p均0.05)。结论马来酸噻吗洛尔可诱导血管内皮细胞凋亡,毛细血管数目减少。马来酸噻吗洛尔与聚桂醇联合应用可起协同作用,可致管腔缩窄,血管壁结构消失,损伤血管内皮并诱导毛细血管内皮细胞发生凋亡,并且不会造成鸡冠外形的改变,符合无创治疗血管瘤要求,为血管瘤的非手术治疗提供理论依据和治疗参考数据。
[Abstract]:Objective to study the histopathological changes and the apoptosis of vascular endothelial cells (VEC) in the model of capillary hemangioma treated with tiamol maleate eye drops. To explore the effect of local wet compress of tiamolol maleate on capillary injury and the feasibility of combined injection of polycinnamol to improve the efficacy of polycinnamol. Methods 30 healthy male chickens aged 6 months were randomly divided into five groups. There were 6 rats in each group. The control group was given 0.9% sodium chloride solution for external use. Experimental group A was treated with tiamolol maleate eyedrops. Group B was treated with triamcinolone acetonide acetate injection and group C was injected with polycinnamol maleate. To observe the general situation of the animal model after drug treatment. A chicken was randomly selected from five ABCDE groups on the 7th day, 14th day, 21st day and 28th day after treatment. The chicken was fixed in 10% formalin solution, stained with HE, and 5 high power field of vision was selected under optical microscope (10 x 40 times). The number of capillaries was counted, the average of capillaries was calculated, and the apoptosis of capillary endothelial cells was observed by TUNEL staining. The number of capillaries was decreased under light microscope, compared with E group. The results showed that the apoptosis of capillary endothelial cells in dermal papilla was observed by TUNEL staining, and a small number of apoptotic capillary endothelial cells were observed in dermal reticular layer of polycinnamol treated group. Moreover, the number of apoptotic cells increased gradually with the prolongation of the observation cycle. There were significant differences in the number of apoptotic capillaries between group E and group A (P < 0.05). Conclusion tiamolol maleate can induce apoptosis of vascular endothelial cells. The combination of thiamolol maleate and polycinnamol may play a synergistic role in constricting the lumen, disappearing the vascular wall structure, damaging the vascular endothelium and inducing the apoptosis of the capillary endothelial cells. And it does not cause the change of the shape of chicken crown, which accords with the requirement of noninvasive treatment of hemangioma, and provides the theoretical basis and treatment reference data for the non-surgical treatment of hemangioma.
