亚硒酸钠对肺癌A549细胞自噬相关蛋白Beclin1、LC3的作用

【摘要】:目的观察亚硒酸钠对肺癌细胞株A549细胞株自噬相关蛋白Beclin1、LC3的作用。方法将人肺癌A549细胞株分为0、0.5、2.5、5.0、8.0μmol/L亚硒酸钠共5组,用不同浓度的亚硒酸钠培养液培养24h、48h后,用免疫细胞化学染色和Western blot印迹法检测亚硒酸钠对自噬相关蛋白Beclin1、LC3表达的影响。结果人肺癌A549细胞株用不同浓度的亚硒酸钠培养24h,48h后,细胞数量逐渐减少,细胞肿胀,变大变圆,染色质凝集、核固缩并断裂。免疫细胞化学和Western blot印迹法结果均显示:随着亚硒酸钠浓度增加,肺癌A549细胞中Beclin1和LC3蛋白免疫细胞化学染色的平均光密度(MOD)有明显提高(P0.05)。结论亚硒酸钠可以抑制肺癌A549细胞生长,诱导肺癌A549细胞自噬。
[Abstract]:Objective to observe the effect of sodium selenite on autophagy associated protein Beclin1 (LC3) in human lung cancer cell line A549. Methods Human lung cancer A549 cell line was divided into 5 groups. The cells were cultured with different concentrations of sodium selenite for 24 h or 48 h. The effect of sodium selenite on the expression of Beclin1 hLC3 was detected by immunocytochemical staining and Western blot blotting. Results after cultured with different concentrations of sodium selenite for 48 h, the number of A549 cells gradually decreased and the cells became swollen. The results of immunocytochemistry and Western blot blotting showed that: with the increase of sodium selenite concentration, The average optical density (mod) of Beclin1 and LC3 protein immunocytochemical staining in lung cancer A549 cells increased significantly. Conclusion Sodium selenite can inhibit the growth of lung cancer A549 cells and induce autophagy of lung cancer A549 cells.
