Notch3、Lgr5和NF-κB在人大肠腺癌组织中的表达及意义

【摘要】:目的:观察Notch3、Lgr5和NF-κB在人大肠腺癌组织、大肠腺瘤组织以及正常大肠黏膜中的表达情况,探讨其与大肠腺癌临床病理特征之间的关系。方法:本实验选择54例大肠腺癌组织、30例大肠腺瘤组织及12例正常大肠黏膜组织,采用免疫组织化学染色SP法检测各组织中Notch3、Lgr5、NF-κB蛋白的表达情况。结合大肠腺癌患者的临床病理资料,比较各组组织中三种蛋白因子的表达与临床病理特征的相关性。结果:Notch3在大肠腺癌、大肠腺瘤、正常大肠黏膜中的表达阳性率分别为63.0%、26.7%、8.3%,癌组与腺瘤组及正常组相比,差异均具有统计学意义(P0.05),腺瘤组与正常组相比,差异无统计学意义(P0.05);Lgr5在大肠腺癌、大肠腺瘤、正常大肠黏膜中的表达阳性率分别为57.4%、36.7%、16.7%,癌组与腺瘤组及正常组相比,差异均具有统计学意义(P0.05),腺瘤组与正常组相比,差异无统计学意义(P0.05);NF-κB在大肠腺癌、大肠腺瘤、正常大肠黏膜中的表达阳性率分别为63.0%、30.0%、25.0%,癌组与腺瘤组及正常组相比,差异均具有统计学意义(P0.05),腺瘤组与正常组相比,差异无统计学意义(P0.05)。Notch3、Lgr5、NF-κB在大肠腺癌中的表达均与性别、肿瘤大小及分化程度无关(P0.05),而均与淋巴结转移及Dukes分期相关(P0.05),伴有淋巴结转移者表达强度高于无淋巴结转移者(P0.05),Dukes分期级别越高,其表达程度越高(P0.05)。此外,Notch3的表达与年龄无关(P0.05),而与肿瘤浸润深度有关,肿瘤浸润深度越大,表达强度越强(P0.05);Lgr5与年龄及肿瘤浸润深度无关(P0.05);NF-κB与肿瘤原发部位无关(P0.05),而与肿瘤浸润深度有关,肿瘤浸润深度越大,表达越强(P0.05);大肠腺癌中Notch3与Lgr5的表达呈正相关(r=0.325,P0.05),Notch3与NF-κB表达无线性相关(P0.05),Lgr5与NF-κB表达无线性相关(P0.05)。结论:1.Notch3、Lgr5、NF-κB在正常大肠黏膜、大肠腺瘤及大肠腺癌组织中的表达呈逐渐增高趋势,三者可能与大肠腺癌的发生具有一定的关系。2.Notch3、Lgr5、NF-κB三者的表达与大肠腺癌的Dukes分期及有无淋巴结转移显著相关,而且Notch3、NF-κB两者还与大肠腺癌的浸润深度显著相关,因此,上述三个指标可在一定程度上预测大肠腺癌的恶性程度及判断患者预后,其三者的高表达也可以作为大肠腺癌淋巴结转移、Dukes分期的重要生物学指标。3.Notch3、Lgr5在大肠腺癌中的表达呈正相关。
[Abstract]:Objective: to observe the expression of Notch3G Lgr5 and NF- 魏 B in human colorectal adenocarcinoma, colorectal adenoma and normal colorectal mucosa. Methods: 54 cases of colorectal adenocarcinoma, 30 cases of colorectal adenoma and 12 cases of normal colorectal mucosa were selected. Immunohistochemical staining SP method was used to detect the expression of Notch3ONg5NF-kappa B protein in various tissues, and combined with clinicopathological data of colorectal adenocarcinoma patients, the expression of NF- 魏 B protein was detected by immunohistochemical staining. Results the positive rates of the expression of the three protein factors in colorectal adenocarcinoma, colorectal adenoma and normal colorectal mucosa were 63.0 and 26.78.3.The positive rate of the tumor group was higher than that of the adenoma group and the normal mucosa. There was no significant difference between adenoma group and normal group. The positive rates of Lgr5 expression in colorectal adenocarcinoma, adenoma and normal mucosa were 57.4% and 36.7%, respectively. The positive rates of NF- 魏 B in adenoma group, adenoma group and normal mucosa were 63.0% and 30.0% respectively. The positive rate of NF- 魏 B in adenoma group was higher than that in adenoma group and normal group, and the positive rates of NF- 魏 B in adenoma group and normal mucosa were 63.0% and 25.0% respectively, and the positive rate of NF- 魏 B in adenoma group was significantly higher than that in normal group. There was no significant difference in the expression of NF- 魏 B between adenoma group and normal group. The tumor size and differentiation degree were not correlated with lymph node metastasis and Dukes stage. The expression intensity of tumor with lymph node metastasis was higher than that without lymph node metastasis. In addition, the expression of Notch3 was not related to age, but was related to the depth of tumor invasion. The stronger the expression intensity was, the more P0.05 Lgr5 was not related to age and depth of tumor invasion. NF- 魏 B was not related to the primary site of the tumor, but was related to the depth of tumor invasion. The expression of Notch3 and Lgr5 in colorectal adenocarcinoma was positively correlated with the expression of NF- 魏 B and the expression of P0.05, Lgr5 and NF- 魏 B.Conclusion the expression of Notch3 and NF- 魏 B in normal colorectal mucosa, colorectal adenoma and adenocarcinoma of large intestine showed a tendency to increase gradually, and the expression of Lgr5NF- 魏 B in normal colorectal mucosa, colorectal adenoma and adenocarcinoma of large intestine showed a tendency of increasing gradually, and the expression of P0.05 and NF- 魏 B in colorectal adenocarcinoma was increased gradually, conclusion: 1.Notch3Lgr5NF- 魏 B in normal colorectal mucosa, colorectal adenoma and adenocarcinoma of large intestine showed an increasing tendency. 2. The expression of NF- 魏 B was significantly correlated with the Dukes stage and lymph node metastasis of colorectal adenocarcinoma, and Notch3NF- 魏 B was significantly correlated with the depth of invasion of colorectal adenocarcinoma. The above three indexes can predict the malignancy of colorectal adenocarcinoma to some extent and judge the prognosis of the patients. The high expression of these three proteins can also be regarded as an important biological index of Dukes staging in colorectal adenocarcinoma. 3. The expression of Notch3Lgr5 in colorectal adenocarcinoma is positively correlated.
