MMP-14和CD31-MVD在结直肠癌组织中的表达及临床意义研究

【摘要】：目的:探讨基质金属蛋白酶-14(matrixmet-alloproteinase-14 MMP-14)和CD31标记的微血管密度(CD31-microvascular density)在结直肠癌组织中的表达及其与肿瘤侵袭转移的临床意义。简要分析MMP-14和CD31-MVD与结直肠癌肿瘤发生、发展以及侵袭转移的相关性,以期为今后结直肠癌患者的治疗以及预后提供一定的指导依据。方法:1.采用免疫组织化学方法检测60例结直肠癌患者的癌组织及20例正常切缘组织中MMP-14和CD31-MVD的表达并分析两者与结直肠癌患者临床病理资料的关系。2.采用q RT-PCR的方法检测20例新鲜结直肠癌组织及正常切缘组织中MMP-14m RNA的相对表达量并分析其与结直肠癌的关系。3.采用SPSS17.0统计软件对数据进行统计分析,结合临床病理资料分析MMP-14和CD31与结直肠肿瘤之间的相关性,并探讨两者的临床意义。结果:1.结直肠癌组织中的MMP-14阳性表达率以及CD31-MVD值明显高于正常切缘组织(P0.05)。2.MMP-14的表达及CD31-MVD值与肿瘤大小、浸润深度、TNM分期和淋巴结转移有相关性(P0.05),而与患者性别、年龄无明显相关(P0.05)。3.MMP-14的表达与CD31-MVD呈正相关(P0.05)。4.结直肠癌组织中MMP-14m RNA的相对表达量显著高于正常切缘组织(P0.01)。结论:结直肠癌组织中MMP-14的高表达和CD31标记的肿瘤组织微血管密度与结直肠癌的发生、浸润、转移有密切关系。检测MMP-14和CD31-MVD值对结直肠癌患者的临床治疗及预后有一定的指导意义。
[Abstract]:Objective: to investigate the expression of matrix metalloproteinase-14 (MMP-14) and microvascular density (CD31-microvascular density) labeled by CD31 in colorectal carcinoma and its clinical significance in tumor invasion and metastasis.To analyze the correlation between MMP-14 and CD31-MVD and the occurrence, development, invasion and metastasis of colorectal cancer, so as to provide some guidance for the treatment and prognosis of colorectal cancer patients in the future.Method 1: 1.Immunohistochemical method was used to detect the expression of MMP-14 and CD31-MVD in 60 cases of colorectal cancer and 20 cases of normal incisor tissues and to analyze the relationship between them and clinicopathological data of colorectal cancer.Q RT-PCR method was used to detect the relative expression of MMP-14m RNA in 20 fresh colorectal cancer tissues and normal incisal margin tissues and to analyze the relationship between MMP-14m RNA and colorectal cancer.The correlation between MMP-14, CD31 and colorectal neoplasms was analyzed by SPSS17.0 software and the clinicopathological data. The clinical significance of the two data was discussed.The result is 1: 1.The positive expression rate of MMP-14 and CD31-MVD in colorectal cancer tissue were significantly higher than those in normal incisal margin tissue (P 0.05N. 2.MMP-14). The expression of CD31-MVD was correlated with tumor size, depth of invasion and lymph node metastasis.There was no significant correlation between age and CD31-MVD.The relative expression of MMP-14m RNA in colorectal cancer was significantly higher than that in normal incisal tissue (P 0.01).Conclusion: the high expression of MMP-14 and the microvessel density of tumor tissue labeled with CD31 are closely related to the occurrence, invasion and metastasis of colorectal carcinoma.The detection of MMP-14 and CD31-MVD values has certain guiding significance for the clinical treatment and prognosis of colorectal cancer patients.
