姜黄素通过上皮-间质转化过程对口腔鳞癌细胞株HN4侵袭转移的影响

【摘要】：目的旨在探讨姜黄素对口腔鳞癌细胞株HN4的增殖、侵袭和凋亡效应及其分子机制。方法 0~60μmol/L姜黄素作用于口腔鳞癌细胞株HN4,24 h后应用MTT法检测细胞的生长活性;划痕实验检测细胞迁移作用;Transwell实验测定对细胞的侵袭作用;流式细胞仪检测细胞凋亡;Western blot测定P53、E-cadherin和Snail蛋白表达水平。结果 MTT检测结果显示各浓度姜黄素对癌细胞生长有抑制,其中,20μmol/L姜黄素有明显抑制。随着姜黄素浓度的增加,对细胞凋亡有明显作用。姜黄素处理后细胞周期生长阻滞于G_0/G_1期。Snail的蛋白表达量随着姜黄素的浓度增加而减少。P53和E-cadherin的表达量随着姜黄素的浓度增加而增加。结论姜黄素能显著抑制口腔鳞癌细胞株HN4的体外生长,上调P53蛋白表达诱导细胞凋亡可能是其作用机制之一。
[Abstract]:Objective to investigate the effect of curcumin on proliferation, invasion and apoptosis of oral squamous cell carcinoma cell line HN4 and its molecular mechanism.Methods #number0# 渭 mol/L curcumin was applied to oral squamous cell carcinoma cell line HN4H for 24 h. The cell growth activity was detected by MTT assay, the cell migration was detected by scratch assay and the invasion effect was determined by Transwell assay.Apoptosis was detected by flow cytometry and the expression of P 53 C-cadherin and Snail protein was detected by Western blot.Results the results of MTT showed that curcumin could inhibit the growth of cancer cells, and 20 渭 mol/L curcumin could inhibit the growth of cancer cells.With the increase of curcumin concentration, it has obvious effect on cell apoptosis.After curcumin treatment, cell cycle growth arrest at G_0/G_1 phase. Snail protein expression decreased with the increase of curcumin concentration, and the expression of. P53 and E-cadherin increased with the increase of curcumin concentration.Conclusion curcumin can significantly inhibit the growth of oral squamous cell carcinoma cell line HN4 in vitro, and upregulation of p53 protein expression may be one of its mechanisms.
