MMP-11蛋白在乳腺癌中的表达与外周血循环肿瘤细胞的相关性分析

【摘要】:目的:研究MMP-11蛋白在乳腺癌中的表达情况以及外周血循环肿瘤细胞在乳腺癌中的临床意义,探讨MMP-11蛋白的表达与乳腺癌外周血循环肿瘤细胞的相关性。方法:收集2015年7月至2015年12月在广西医科大学第一附属医院胃肠腺体二病区新确诊为乳腺癌并且未经过任何手术、放疗、化疗治疗的患者137例。在手术前采集患者外周血10ml,运用Canpatrol~(TM) CTCs方法检测乳腺癌外周血循环肿瘤细胞,运用免疫组化方法检测MMP-11蛋白在乳腺癌中的表达情况。结果:137例入组病例,MMP-11蛋白在100例乳腺癌组织中表达阳性,37例表达阴性,阳性率为72%;MMP-11蛋白的表达与乳腺癌患者临床分期具有相关性(P=0.049);137例乳腺癌患者中112例乳腺癌患者外周血检测出循环肿瘤细胞,阳性率为82%;在137例患者中,总CTCs数量平均数为4.73(0-36),上皮型CTCs数量平均数为1.99(0-22),混合型CTCs为2.06(0-17),间质型CTCs为0.69(0-7);在112例CTCs阳性患者中,三种类型CTCs所占CTCs比例的平均值:上皮型CTCs为48.7%,混合型CTCs为39.8%,间质型CTCs为11.5%;乳腺癌外周血总CTCs的阳性检测结果与MMP-11蛋白在乳腺癌中的表达具有相关性(P=0.009);总CTCs在数量上与MMP-11蛋白表达具有相关性(P=0.042)。结论:MMP-11在乳腺癌中的表达与循环肿瘤细胞具有相关性,MMP-11蛋白可能通过改变乳腺癌肿瘤微环境促进循环肿瘤细胞的形成;Canpatrol~(TM) CTCs循环肿瘤细胞检测技术检测乳腺癌外周血循环肿瘤细胞阳性率为82%,Canpatrol~(TM) CTCs检测技术是一种高效CTCs检测方法。
[Abstract]:Objective: to study the expression of MMP-11 protein in breast cancer and the clinical significance of circulating tumor cells in peripheral blood, and to explore the correlation between the expression of MMP-11 protein and circulating tumor cells in peripheral blood of breast cancer.Methods: from July 2015 to December 2015, 137 patients with breast cancer who were newly diagnosed as breast cancer in the second section of gastrointestinal gland of the first affiliated Hospital of Guangxi Medical University without any operation, radiotherapy and chemotherapy were collected.The peripheral blood of 10 ml patients was collected before operation. The circulating tumor cells in peripheral blood of breast cancer were detected by Canpatrola CTCs method and the expression of MMP-11 protein in breast cancer was detected by immunohistochemical method.Results the positive expression of MMP-11 protein in 100 cases of breast cancer was negative in 37 cases.The positive rate of MMP-11 protein expression was correlated with clinical stage of breast cancer patients. Circulating tumor cells were detected in peripheral blood of 112 breast cancer patients out of 137 breast cancer patients, and the positive rate was 82% in 137 breast cancer patients.The average number of total CTCs, epithelial CTCs, mixed CTCs and interstitial CTCs were 4.73 ~ 0 ~ 36, 1.99 ~ 22 ~ 22, 2.06 ~ 17 ~ 17 and 0.69 ~ 0 ~ 7, respectively.The average proportion of CTCs in three types of CTCs was 48.7 in epithelial type, 39.8 in mixed type and 11.5 in interstitial CTCs. The positive results of total CTCs in peripheral blood of breast cancer were correlated with the expression of MMP-11 protein in breast cancer.There was a correlation between the quantity and the expression of MMP-11 protein.Conclusion the expression of MMP 11 in breast cancer is associated with circulating tumor cells. The expression of MMP 11 protein may promote the formation of circulating tumor cells by changing the microenvironment of breast cancer. (CTCs) circulating tumor cell detection technique may be used to detect the peripheral tumor cells of breast cancer.The positive rate of tumor cells in blood circulation is 82%. The technique of CTCs is an effective method for detecting CTCs.
