Nek8、BICD2、VHL在结直肠癌组织中的表达及临床病理意义

【摘要】:目的:探讨结直肠癌(colorectal cancer,CRC)组织中Nek8的mRNA表达情况,Nek8、BICD2、VHL的蛋白表达变化及三者与CRC临床病理特征和预后的关系。方法:采用RT-PCR(Reverse Transcription Polymerase Chain Reaction)和RT-qPCR(Real-time RT-PCR)的方法检测30例配对的CRC组织、相应的邻近CRC组织的癌旁组织(距离肿瘤3.0cm处)和相应的邻近CRC组织的正常结直肠黏膜组织(距离肿瘤组织8.0-10.0cm处)中Nek8的mRNA表达情况。应用免疫组织化学的方法(SP法)检测30例配对的CRC组织和相应的邻近CRC组织的正常结直肠黏膜组织中Nek8、BICD2、VHL蛋白表达情况和病理学特征,同时检测20例结直肠黏膜慢性炎症、20例结直肠管状腺瘤(II级)和60例结直肠管状腺癌组织中Nek8、BICD2、VHL蛋白表达情况及与CRC临床病理参数的关系。结果:(1)Nek8 mRNA在CRC组织和癌旁组织中的表达高于结直肠正常黏膜上皮中的表达,其中CRC组织中的表达最高。(2)免疫组化结果显示:30例配对标本中Nek8、BICD2蛋白均在CRC组织中高表达[56.7%(17?30例)、76.7%(23?30例)]且显著高于结直肠正常黏膜组织中的表达[13.3%(4?30例)、13.3%(4?30例)],VHL蛋白在CRC组织中的表达[30.0%(9?30例)]显著低于结直肠正常黏膜组织[63.3%(19?30例)](p0.05);(3)Nek8和BICD2蛋白在结直肠腺瘤组织[80.0%(16?20例)、75.0%(15?20例)]及腺癌组织[83.3%(50?60例)、73.3%(44?60例)]中的表达量明显高于结直肠黏膜慢性炎症组织[25.0%(5?20例)、20.0%(4?20例)],VHL蛋白分别在结直肠黏膜慢性炎症组织、腺瘤组织和腺癌组织中的阳性表达量[75.0%(15?20例)、40.0%(8?20例)、26.7%(16?60例)]呈逐渐降低的趋势(p0.05),三种蛋白的表达分别与年龄、性别、浸润深度、淋巴结转移、临床分期、五年生存率、远处转移无关(p0.05)。(4)对CRC组织中Nek8、BICD2、VHL分别进行两两比较,Nek8和BICD2两者之间相关性显著,呈正相关的关系(r=0.337,P0.05),余两两之间相关性比较均无统计学意义(p0.05)。结论:Nek8、BICD2在CRC组织中表达增加,VHL在CRC组织中表达减少,三者可能在CRC的发生、发展中起重要的作用。
[Abstract]:Objective: to investigate the expression of Nek8 mRNA in colorectal carcinoma and its relationship with clinicopathological features and prognosis of CRC. Methods: 30 matched CRC tissues were detected by RT-PCR(Reverse Transcription Polymerase Chain reaction and RT-qPCR(Real-time RT-PCR. The expression of Nek8 mRNA in adjacent adjacent tissues of CRC (adjacent to tumor 3.0cm) and normal colorectal mucosa adjacent to CRC (8.0-10.0cm) was observed. Immunohistochemical method was used to detect the expression and pathological features of Nek8 BICD2VHL protein in 30 matched CRC tissues and corresponding normal colorectal mucosa tissues adjacent to CRC. At the same time, the expression of Nek8 BICD2VHL protein and its relationship with clinicopathological parameters of CRC were detected in 20 cases of chronic colorectal mucosal inflammation and 20 cases of colorectal tubular adenoma grade II) and 60 cases of colorectal tubular adenocarcinoma. Results the expression of Nek8 mRNA in CRC and adjacent tissues was higher than that in normal colorectal mucosal epithelium. The expression of Nek8 BICD2 protein in 30 matched specimens of CRC was higher than that in normal colorectal mucosa tissues [56.7 ~ 1730 cases] and was significantly higher than that in normal colorectal mucosa tissue [13.3330 cases] and the expression of Nek8BICD2 protein was significantly higher than that in normal colorectal mucosa tissue [13.3330 cases [13.330 cases].] VHL protein was expressed in the tissues of CRC tissues (56.70.1730 cases), and was significantly higher than that in normal colorectal mucosa tissues (13.3330 cases (13.330 cases)). The expression of CRC in colorectal adenoma [30.030 cases] was significantly lower than that in normal colorectal mucosal tissue [63.33,1930 cases] p0.05 ~ (3) Nek8 and BICD2 protein in colorectal adenoma [80.020 cases of rectal adenoma [80.020 cases] and adenocarcinoma (83.335060 cases of "73.3")] and the expression of BICD2 protein was significantly higher than that of "chronic colorectal mucosal inflammation" (83.335060 cases). The VHL protein was found in the chronic inflammation of colorectal mucosa in 20 cases (20 cases) and 20 cases (20 cases). The positive expression level in adenoma tissue and adenocarcinoma tissue [75.0% 1520 cases (40.0%) and 820 cases (26.7% 1660 cases)] showed a decreasing trend. The expression of three proteins was correlated with age, sex, depth of invasion, lymph node metastasis, clinical stage, 5-year survival rate, respectively. There was significant correlation between Nek8 and BICD2 in CRC tissues. There was a positive correlation between Nek8 and BICD2VHL. There was a positive correlation between Nek8 and BICD2VHL, and there was no significant correlation between the other two groups. Conclusion the expression of BICD2 in CRC tissues increased and decreased in CRC tissues. The expression of BICD2 may play an important role in the occurrence and development of CRC.
