人结直肠癌组织中TLR-4与PGE-2的表达及意义

【摘要】:目的:研究分析Toll样受体-4(toll-like receptor 4,TLR-4)和前列腺素E2(prostaglandin E2,PGE-2)在结直肠癌组织中的表达情况,探讨TLR-4和PGE-2与结直肠癌之间的相关性,对TLR-4相关信号通路的深入研究,有助于我们更好的了解消化道恶性肿瘤的发生机制,TLR-4信号通路有望成为防治消化道肿瘤的新靶点。方法:应用免疫组织化学方法(S-P)法检测40例广西中医药大学患者结直肠癌、癌旁组织和远癌组织中TLR-4、PGE-2的表达。采用SPSS17.0统计学软件分析处理,采用χ2检验,采用Spearman等级相关分析法分析TLR-4与PGE-2表达的相关性,均以P0.05为差异有统计学意义。结果:数据显示TLR-4在远癌结直肠组织上呈微弱表达,在癌旁组织和癌组织中呈阳性表达,且在癌肿瘤中表达明显增强,呈散在游离或颗粒状分布于细胞膜和细胞质上,其定位以胞浆胞膜为主,细胞核上无表达。PGE-2在远癌结直肠组织中呈微弱表达,在癌旁组织及癌组织中的表达明显逐渐增强,其定位也以胞浆胞膜为主,细胞核上亦无表达。TLR-4在癌组织、癌旁组织及远癌组织中的阳性面积率依次下降,分别为9.94%,5.85%,1.30%,三者相比较差异有统计学意义(P0.01)。PGE-2在结直肠癌组织、癌旁和远癌组织中,其表达水平逐渐下调,阳性率分别为3.89%,3.19%,1.01%三者相比较差异有统计学意义(P0.01)。TLR-4在PGE-2阳性组中的表达率为(20/32)62.50%,PGE-2表达率为(4/8)50.00%在阴性组中;PGE-2在TLR-4阳性组中的表达率为(20/24)83.33%,在TLR-4阴性组中的表达率为(12/16)75.00%;在40例结直肠癌组织中,有20例两者同为阳性表达,4例同为阴性表达,表达一致率为60.00%,分析指出两者表达呈正相关,差异有统计学意义(r=0.296,P0.05)。结论:1、TLR-4在结直肠癌组织、癌旁组织和远癌正常组织中的表达水平逐渐下降,距离癌组织中心越远,其表达阳性面积率越低;2、PGE-2在结直肠癌组织、癌旁组织和远癌正常组织中的表达水平逐渐下降,距离癌组织中心越远,其表达阳性面积率越低;3、结直肠癌组织中TLR-4和PGE-2的表达呈正相关,TLR-4可能通过上调PGE-2的表达促进结直肠癌的恶性进展。
[Abstract]:Objective: to investigate the expression of Toll like receptor -4toll-like receptor 4 (TLR-4) and prostaglandin E2(prostaglandin E2P PGE-2 in colorectal cancer, to explore the relationship between TLR-4 and PGE-2 and colorectal cancer, and to investigate the TLR-4 related signal pathway. TLR-4 signaling pathway may be a new target for the prevention and treatment of gastrointestinal cancer. Methods: immunohistochemical method was used to detect the expression of TLR-4 and PGE-2 in 40 patients with colorectal cancer, paracancerous tissues and distant carcinomas in Guangxi University of traditional Chinese Medicine. The correlation between TLR-4 and PGE-2 expression was analyzed by SPSS17.0 statistical software, 蠂 2 test and Spearman rank correlation analysis. Results: the data showed that the expression of TLR-4 was weakly expressed in distal colorectal tissues, positive in paracancerous tissues and cancerous tissues, and increased in cancer tumors, and distributed in free or granular form on cell membrane and cytoplasm. The localization of PGE-2 was mainly in the cytoplasmic membrane, and the expression of PGE-2 was weak in the distal colorectal tissues, and increased gradually in the adjacent tissues and cancer tissues. The localization of PGE-2 was mainly in the cytosolic membrane. The positive area of TLR-4 in cancer tissues, paracancerous tissues and distal carcinomas decreased in turn, 9.945.85 and 1.30. the difference was statistically significant in colorectal cancer tissues, paracancerous tissues and distal cancer tissues, and the difference was significant in colorectal cancer tissues, paracancerous tissues and distal cancer tissues. The positive rate of TLR-4 in cancer tissue, paracancerous tissue and distal cancer tissue was significantly lower than that in the control group (P < 0.05). Its expression level was gradually down-regulated. The positive rate of PGE-2 was 3.89% 3.19% and 1.01%, respectively. There was a significant difference among the three groups. The expression rate of P0.01U. TLR-4 in the PGE-2 positive group was 20 / 32, 62.50% and 50.00% respectively. In the negative group, the expression rate of PGE-2 in the TLR-4 positive group was 20 / 2483.330.The expression rate in the TLR-4 negative group was 12 / 16 / 75.00, and the positive rate of PGE-2 in the negative TLR-4 group was 20 / 2483.330.The expression rate of PGE-2 in the TLR-4 negative group was 12 / 16 / 75.00; In colorectal cancer, There were 20 cases of both positive expression and 4 cases of negative expression, the consistent rate of expression was 60.00. the analysis showed that the expression of both positive correlation, the difference was statistically significant (P 0.05). Conclusion the expression of TLR-4 in colorectal cancer tissues, paracancerous tissues and normal tissues of distal carcinoma decreased gradually. The farther away from the center of cancer tissue, the lower the positive area of TLR-4 expression was in colorectal cancer tissues. The level of expression in paracancerous tissues and normal tissues of distant cancer decreased gradually, and the farther away from the center of cancer tissue, The lower the positive area rate of TLR-4, the higher the expression of TLR-4 and PGE-2 in colorectal cancer tissues. TLR-4 may promote the malignant progression of colorectal cancer by up-regulating the expression of PGE-2.
