miR-126、miR-355及Exportin-5在肺癌中表达研究

【摘要】:目的:探讨miR-126、miR-355及Exportin-5在肺癌中表达及相互之间关系。方法:收集47例肺癌患者的癌组织及癌旁组织,利用TRIzol提取总RNA,使用定时荧光定量PCR检测基因的相对表达量。结果:肺癌组织与癌旁组织中的miR-126,Exportin-5差异具有统计学意义(P0.05),癌旁组织中miR-126与pri-miR-126(r=-0.31)呈负相关(P0.05),癌组织中基因表达量与临床指标之间的关系中发现miR-126与MRP(r=0.43)呈正相关(P0.05),Exportin-5与TOPO(r=-0.36)、pri-miR-126与饮酒(r=-0.34)、MRP(r=-0.43)呈负相关(P0.05)。结论:miR-126、Exportin-5在肺癌组织中表达下调,基因在肺癌发生发展过程中可能通过抑制基因的表达促进肺癌的发生与发展。
[Abstract]:Objective: to investigate the expression and relationship of miR-126 miR-355 and Exportin-5 in lung cancer. Methods: the cancer tissues and adjacent tissues of 47 patients with lung cancer were collected. The total RNAs were extracted by TRIzol and the relative expression of genes was detected by timed fluorescence quantitative PCR. Results: the difference of miR-126 exportin-5 between lung cancer and paracancerous tissues was statistically significant (P 0.05), and the expression of miR-126 was negatively correlated with pri-miR-126 r-0.31 (P 0.05). A positive correlation was found between miR-126 and MRPr0.43). There was a negative correlation with the alcohol consumption (r = 0.34) and MRP (r = 0.43) (P = 0.05, P = 0.05). Conclusion the expression of 1: miR-126 exportin-5 is down-regulated in lung cancer tissues, and the gene may promote the occurrence and development of lung cancer by inhibiting the expression of genes during the carcinogenesis and development of lung cancer.
