青蒿琥酯对人结肠癌细胞HCT116和HT29增殖与凋亡的影响

【摘要】:目的:观察青蒿琥酯对人结肠癌细胞HCT116和HT29抑制和凋亡的影响,为日后临床结直肠癌(CRC)的治疗提供可行的新的辅助化疗治疗手段。方法:用不同浓度青蒿琥酯处理两种人结肠癌细胞HCT116和HT29,应用四甲基偶氮唑蓝(MTT)比色法在酶标仪490nm波长处测量每孔的吸光度(A)值,检测青蒿琥酯对人结肠癌细胞HCT116和HT29的抑制效应;应用流式细胞术(FCM)检测人结肠癌细胞HCT116和HT29细胞凋亡的情况。使用SPSS13.0统计软件处理数据,实验数据(即细胞抑制率和凋亡率)采用于均数±标准差(`X±s)表示,HCT116和HT29抑制率与凋亡率组内两两比较应用两样本t检验,组内抑制率和凋亡率的比较分别用单因素方差分析和最小差异t测试(LSD-t)分析、浓度与抑制率两者关系采用双变量线性相关分析。P0.05,差异具有统计学意义。结果:不同浓度青蒿琥酯(7.5、15、30、60、120μg·ml-1)处理两种人结肠癌细胞48 h后,抑制率随浓度增加呈上升趋势(HCT116:19.42%、24.59%、44.84%、74.15%、92.87%;HT29:11.77%、17.63%、34.23%、48.51%、70.59%);且同种浓度下,对HCT116作用明显强于HT29。不同浓度青蒿琥酯(0、60、120μg·ml-1)处理两种人结肠癌细胞24 h后凋亡率分别为(HCT116:2.46%51.36%64.10%;HT29:1.14%35.95%43.45%),加药组与对照组、同浓度下两种细胞凋亡均有统计学意义。结论:青蒿琥酯对人结肠癌细胞HCT116和HT29具有生长抑制作用,与浓度曾正相关作用,且对HCT116作用强于HT29;青蒿琥酯对人结肠癌细胞HCT116和HT29具有诱导凋亡作用,且对HCT116作用强于HT29。
[Abstract]:Aim: to observe the effect of artesunate on the inhibition and apoptosis of HCT116 and HT29 in human colon cancer cells, and to provide a new adjuvant chemotherapy for the treatment of colorectal cancer. Methods: two kinds of human colon cancer cell lines HCT116 and HT29 were treated with artesunate of different concentrations. The absorbance of each hole was measured by MTT colorimetry at the wavelength of 490nm. The inhibitory effect of artesunate on HCT116 and HT29 in human colon cancer cells and apoptosis in HCT116 and HT29 cells were detected by flow cytometry. SPSS13.0 statistical software was used to process the data. The experimental data (that is, cell inhibition rate and apoptosis rate) were measured by mean 卤standard deviation (`X 卤s). The inhibition rates of HCT116 and HT29 were compared with those in the apoptotic rate group. Two samples t test were used to compare the inhibition rate of HCT116 and HT29. Intra-group inhibition rate and apoptosis rate were analyzed by single factor ANOVA and least difference t test (LSD-t). The relationship between concentration and inhibition rate was analyzed by bivariate linear correlation analysis. P0.05. the difference was statistically significant. Results: after treated with different concentrations of artesunate 7.51530C 60120 渭 g ml-1 for 48 h, the inhibition rate of HCT116: 19.42C24.5959and HCT116: 19.42ML-1 was higher than that of HT292911.7717.637.2378.51.The inhibitory rate of HCT116: 19.42C24.5945 was better than that of HT299.The inhibition rate of HCT116: 19.42 + 24.59mg / ml was much stronger than that of HT299.The inhibitory effect of HCT116: 19.42C24.590.59 was better than that of HT29. Two kinds of human colon cancer cells were treated with different concentrations of artesunate (60120 渭 g ml-1) for 24 hours. The apoptotic rates of two kinds of human colon cancer cells were as follows: HCT116: 2.46Q.36d.10and HT29: 1.145.95C.450.There were significant differences between the two groups. Conclusion: artesunate can inhibit the growth of human colon cancer cell line HCT116 and HT29, which is positively related to the concentration of artesunate, and has stronger effect on HCT116 than HT29.The artesunate can induce apoptosis in human colon cancer cell line HCT116 and HT29, and the effect on HCT116 is stronger than that on HCT116.
