【摘要】:背景p-21活化激酶(PAK)是小鸟苷三磷酸酶的下游靶蛋白,在许多基本的细胞功能中发挥着重要的作用,包括细胞骨架重组、细胞运动。PAK在多种组织中广泛表达,在多种类型的癌症中过度表达。本研究的目的是探讨PAK1与PAK4与结直肠癌临床病理特征的关系。方法 PAK1和PAK4在结直肠肿瘤患者的高度表达是通过RT-PCR、免疫组化进行临床分析。结果PAK1与PAK4基因在结直肠癌组织中的相对表达水平明显高于正常组织(P0.01)。 PAK4表达明显高于在同一癌组织中的PAK1。PAK1和PAK4表达随癌组织的临床分期增加而逐渐升高(P0.01)。PAK1在淋巴结阳性的患者中高表达,PAK4在浸润浆膜层患者中高表达(P0.05)。PAK1高表达组与PAK1低表达组相比有较高的复发转移率。后续分析显示,中位无进展生存时间的PAK1高表达组较Pakl的低表达组明显缩短。结论PAK1与PAK4的表达与结直肠癌的浸润和转移相关,PAK1的高表达可能表明大肠癌预后不良。
[Abstract]:Background p-21 activated kinase (pak) is a downstream target protein of guanosine triphosphatase, which plays an important role in many basic cellular functions, including cytoskeleton recombination, cell motility, PAK expression in a variety of tissues. Overexpression in many types of cancer. The purpose of this study was to investigate the relationship between PAK1 and PAK4 and clinicopathological features of colorectal cancer. Methods the high expression of PAK1 and PAK4 in colorectal cancer patients was analyzed by RT-PCR and immunohistochemistry. Results the relative expression levels of PAK1 and PAK4 genes in colorectal carcinoma were significantly higher than those in normal tissues (P 0.01). The expression of PAK4 was significantly higher than that of PAK1.PAK1 and PAK4 in the same cancer tissue. With the increase of clinical stage of cancer tissue, the expression of p0. 01 and PAK1 in lymph node positive patients was significantly higher. The expression of p0. 05 and PAK1 in infiltrating serous layer patients was higher than that in the high expression group of P0. 05 and PAK1. The recurrence and metastasis rate of low expression group was higher than that of low expression group. The follow-up analysis showed that the high expression group of PAK1 with median survival time without progression was significantly shorter than the group with low expression of Pakl. Conclusion the expression of PAK1 and PAK4 may be associated with the invasion and metastasis of colorectal cancer and the high expression of PAK1 may indicate that the prognosis of colorectal cancer is poor.
