注射用黄芪多糖体外诱导人鼻咽癌CNE-2细胞自噬及其机制

【摘要】:目的:探讨注射用黄芪多糖诱导人鼻咽癌细胞株CNE-2的自噬作用。方法:采用CCK8检测注射用黄芪多糖对CNE-2增殖的抑制作用;吖啶橙染色观察注射用黄芪多糖作用于CNE-2细胞引起的自噬形态变化;透射电镜观察注射用黄芪多糖作用于CNE-2细胞后引起的细胞超微结构变化。Western blot实验检测注射用黄芪多糖作用于CNE-2细胞后Beclin-1的表达变化。结果:注射用黄芪多糖对CNE-2细胞的生长有明显抑制作用,且具有浓度及时间依赖性。注射用黄芪多糖作用后可诱导CNE-2细胞发生自噬。透射电镜下细胞体内大空泡及膜性双层结构增多;吖啶橙染色可见注射用黄芪多糖作用后观察到更多的亮红色酸性自噬泡。Western blot结果表明Beclin-1蛋白表达增高。结论:注射用黄芪多糖抑制细胞增殖,可能通过上调beclin1表达诱导人鼻咽癌细胞CNE-2细胞发生自噬。
[Abstract]:Objective: To investigate the autophagy effect of injection of Astragalus Polysaccharides on human nasopharyngeal carcinoma cell line CNE-2. Methods: CCK8 was used to detect the inhibitory effect of Astragalus Polysaccharides on CNE-2 proliferation; acridine orange staining was used to observe the changes of autophagy induced by Astragalus Polysaccharide on CNE-2 cells. The transmission electron microscope was used to observe the effect of Astragalus Polysaccharide on CNE-2 The ultrastructural change of cells after the cell.Western blot test was used to detect the expression of Beclin-1 in CNE-2 cells after injection of Astragalus polysaccharides. Results: the injection of Astragalus Polysaccharides had obvious inhibitory effect on the growth of CNE-2 cells, and was dependent on the concentration and time. The injection of Astragalus polysaccharides could induce the occurrence of CNE-2 cells. Autophagy. Under transmission electron microscope, the large vacuoles and membranous double layer structures in the cells in the cells increased; acridine orange staining showed that more bright red acid autophagic vesicles.Western blot results showed that the expression of Beclin-1 protein increased after injection of Astragalus polysaccharides. Conclusion: injection of Astragalus polysaccharide to inhibit cell proliferation may be induced by up regulation of Beclin1 expression. Autophagy occurs in the CNE-2 cells of human nasopharyngeal carcinoma cells.
