HDAC1、HDAC5和E-cadherin在肺腺癌中的表达和相关性研究

【摘要】:【目的】采用免疫组织化学的方法,在肺腺癌癌组织和癌旁组织中,分别对HDAC1、HDAC5和E-cadherin三种蛋白的表达水平进行检测,同时对HDAC1、HDAC5和E-cadherin在肺腺癌癌组织和癌旁组织中的表达情况进行对比分析,探讨三种蛋白在肺腺癌组织中的表达情况与各临床病理特征之间的关系以及三者之间共同表达的情况,为肺腺癌临床个体化治疗及寻找新的靶点提供相关理论依据。【方法】应用免疫组化SP法检测在93例肺腺癌组织和48例癌旁正常肺组织中应用免疫组织化学SP法分别检测HDAC1、HDAC5及E-cadherin的表达结果。采用χ2检验和四格表精确概率法分析HDAC1、HDAC5和E-cadherin的表达情况以及三种蛋白与肺腺癌中的变大情况与各临床病理特征之间的关系。采用Spearman相关性分析对三种蛋白之间的相关性进行统计学分析,所有实验所得数据均在SPSS 21.0统计软件上进行处理。P0.05则表明差异有统计学意义。【结果】在93例肺腺癌患者的组织中,HDAC1、HDAC5和E-cadherin的阳性表达率分别为58.06%(54/93),48.39%(45/93),45.16%(42/93),在48例癌旁组织中,HDAC1、HDAC5和E-cadherin的阳性表达率分别为20.83%(10/48),66.67%(32/48),68.75%(33/48),三种蛋白在肺腺癌组织与癌旁组织中的阳性表达率有显著性差异(P0.05)。E-cadherin的表达与年龄及细胞分化程度密切相关(P0.05),与其他病理特征无明显相关,HDAC1的表达与年龄密切相关(P0.05),与其他病理特征无明显相关,HDAC5与临床病理特征无明显相关(P0.05)。HDAC1与HDAC5、HDAC1与E-cadherin之间呈负相关(r分别为-0.224、-0.236,P0.05),HDAC5与E-cadherin之间无明显相关性(r=0.159,,P=0.128)。【结论】肺腺癌的发生发展是多因素、多步骤、多环节共同参与作用的复杂结果, HDAC1与HDAC5、HDAC1与E-cadherin之间呈负相关,HDAC5与E-cadherin之间无明显相关性。HDAC1的表达与年龄密切相关,E-cadherin的表达与年龄及细胞分化程度密切相关,联合检测三者可能对判断预后、合理选择药物有一定指导意义。
[Abstract]:[objective] to detect the expression of HDAC1HDAC5 and E-cadherin in lung adenocarcinoma tissues and adjacent tissues by immunohistochemical method. At the same time, the expression of HDAC5 and E-cadherin in lung adenocarcinoma tissues and adjacent tissues were compared and analyzed, and the relationship between the expression of HDAC5 and E-cadherin in lung adenocarcinoma tissues and the clinicopathological features and the co-expression of the three proteins were discussed. [methods] Immunohistochemical SP method was used to detect 93 cases of lung adenocarcinoma and 48 cases of adjacent normal lung tissues. The expression of HDAC5 and E-cadherin were detected. The expression of HDAC5 and E-cadherin was analyzed by 蠂 2 test and quad table exact probability method. The relationship between the expression of HDAC5 and HDAC5 and the relationship between the three proteins and the enlargement of lung adenocarcinoma and the clinicopathological features were analyzed. The correlation between the three proteins was statistically analyzed by Spearman correlation analysis. All experimental data were processed on SPSS 21.0 statistical software. P05 showed significant difference. [results] in 93 cases of lung adenocarcinoma, the positive rates of HDAC5 and E-cadherin were 58.06% 58.06% 48.39% 48.39% 48.39% 43.16% 42.93% and 48 cases of paracancerous tissues were 42.93%, 58.06%, 48.39% and 48.39%, respectively, and in 48 cases of adjacent tissues, the positive rates of HDAC5 and E-cadherin were 58.06%, 48.39% and 48.39%, respectively. The positive expression rates of HDAC5 and E-cadherin were 20.83 / 10 / 48 / 66.67 / 32 / 48 and 68.7575 / 48, respectively. There was a significant difference between the expression of three proteins in lung adenocarcinoma and adjacent tissues. The expression of E-cadherin was closely related to age and cell differentiation (P0.05), but not to other pathological features. There was no significant correlation between HDAC5 expression and clinicopathological features (P 0.05). HDAC1 was negatively correlated with E-cadherin (r = -0.224- 0.236P0.05HDAC5 and E-cadherin). [conclusion] there was no significant correlation between HDAC5 and E-cadherin. [conclusion] there is no significant correlation between HDAC5 and HDAC5 and E-cadherin. [conclusion] The occurrence and development of lung adenocarcinoma are many factors. There was no significant correlation between HDAC1 and HDAC5 and E-cadherin. There was no significant correlation between HDAC5 and E-cadherin. The expression of E-cadherin was closely related to age and the degree of cell differentiation. Combined detection of the three may have certain guiding significance in judging prognosis and selecting drugs reasonably.
