NK细胞对非小细胞肺癌细胞NCI-H292的杀伤作用及对其β-catenin蛋白表达的影响

【摘要】:目的探讨自然杀伤(NK)细胞对肺癌细胞系NCI-H292的杀伤作用及对癌细胞中β-catenin蛋白表达的影响。方法无菌采集健康人外周血,培养NK细胞(NK组),取培养14 d后的NK细胞,调整细胞浓度为5×106/m L,按1∶5的比例接种到NCI-H292细胞(NK+NCI-H292组)培养板共培养24 h,单独培养NCI-H292细胞(NCIH292组)24 h。采用CCK-8法测算NK细胞对肺癌细胞系NCI-H292的杀伤活性,采用Western blotting法检测NCIH292细胞中β-catenin蛋白的表达。结果 NK细胞对NCI-H292细胞的杀伤率为96.720%±1.081%,与NK及NCI-H292组比较,NK+NCI-H292组β-catenin表达降低(P均0.01)。结论 NK细胞对NCI-H292细胞具有杀伤力,同时能够抑制NCI-H292细胞中β-catenin蛋白的表达。
[Abstract]:Objective to investigate the cytotoxicity of natural killer (NK) cells to lung cancer cell line NCI-H292 and the expression of 尾 -catenin in human lung cancer cells. Methods Peripheral blood samples from healthy people were collected and NK cells were cultured in NK group. The NK cells were cultured for 14 days. The concentration of NK cells was adjusted to 5 脳 10 6 / mL. The NK cells were inoculated into the NCI-H292 cell / NK NCI-H292 group at 1:5 ratio for 24 hours. The NCI-H292 cells were cultured alone in NCIH292 group for 24 hours. The cytotoxicity of NK cells to NCI-H292 was measured by CCK-8 assay, and the expression of 尾 -catenin protein in NCIH292 cells was detected by Western blotting assay. Results the killing rate of NK cells on NCI-H292 cells was 96.720% 卤1.081%. Compared with NK and NCI-H292 groups, the expression of 尾 -catenin in NK NCI-H292 group was lower than that in NK NCI-H292 group (P < 0.01). Conclusion NK cells can kill NCI-H292 cells and inhibit the expression of 尾 -catenin in NCI-H292 cells.
