Rock2和Wnt11在食管癌细胞系中的表达及意义

【摘要】:目的早期食管癌临床症状常不明显,肿瘤标志物的深入研究对食管癌早期诊断极为重要。文中通过研究体外特定细胞株[Eca-109及人食管正常上皮细胞(HEEC)]中Rock2和Wnt11蛋白表达量,分析各蛋白的表达与食管癌的关系。方法培养Eca-109和HEEC细胞株,RT-q PCR和Western blot技术检测Rock2和Wnt11在细胞株中的表达水平。结果RT-q PCR检测结果显示,与HEEC相比,Eca-109中Rock2 mRNA、Wnt11 mRNA表达水平明显升高[(1.000±0.000)vs(4.955±0.539),(1.000±0.000)vs(2.925±0.230),P0.01]。Western blot检测显示Rock2和Wnt11蛋白在Eca-109中的表达较HEEC均升高[(955.000±21.628)vs(778.844±102.193),(2 175.316±145.623)vs(1 312.233±50.734),P0.05]。结论 Rock2和Wnt11表达增高可能是食管鳞状细胞癌转移的标志。
[Abstract]:Objective the clinical symptoms of early esophageal carcinoma are not obvious, and the further study of tumor markers is very important for early diagnosis of esophageal carcinoma. In this paper, the expression of Rock2 and Wnt11 proteins in specific cell lines [Eca-109 and human normal esophageal epithelial cells] in vitro was studied, and the relationship between the expression of these proteins and esophageal carcinoma was analyzed. Methods the expression levels of Rock2 and Wnt11 in Eca-109 and HEEC cell lines were detected by RT-q PCR and Western blot techniques. Results compared with HEEC, the expression of Rock2 mRNA-Wnt11 mRNA in Eca-109 was significantly increased [1.000 卤0.000)vs(4.955 卤0.539 P0.01]. Western blot analysis showed that the expression of Rock2 and Wnt11 protein in Eca-109 was significantly higher than that in HEEC [955.000 卤21.628)vs(778.844 卤102.1931] 2 175.316 卤145.623)vs(1 312.233 卤50.734 (P0.05). Conclusion the increased expression of Rock2 and Wnt11 may be a marker for metastasis of esophageal squamous cell carcinoma.
