姜黄素联合顺铂对人舌鳞癌Tca-8113细胞的影响

【摘要】:目的姜黄素或姜黄素联合顺铂在体外作用于人舌鳞癌Tca-8113细胞,观察其对人舌鳞癌细胞增殖的影响,并探讨可能的抗肿瘤作用机制。方法将培养的Tca-8113细胞随机分为姜黄素组、顺铂组、姜黄素联合顺铂组和对照组。MTT法检测各组药物对舌鳞癌Tca-8113细胞增殖的抑制作用;Hoechst 33258染色法观察各组药物对舌鳞癌细胞核的形态影响;流式细胞术检测细胞凋亡率;Western blotting检测各组药物对舌鳞癌Tca-8113细胞Notch1和表皮生长因子受体(EGFR)蛋白表达的影响。结果 MTT法结果显示,姜黄素联合顺铂组抑制舌鳞癌Tca-8113细胞增殖的作用明显高于姜黄素组和顺铂组(P0.05)。流式细胞分析结果显示,姜黄素联合顺铂组的细胞凋亡率明显高于姜黄素组和顺铂组(P0.05)。Western blotting结果显示,与姜黄素联合顺铂组比较,姜黄素组和顺铂组Notch1表达明显上调,EGFR表达则明显下调,而姜黄素联合顺铂组对Notch1和EGFR表达调控的影响与姜黄素组和顺铂组相比差异有统计学意义(P0.05)。结论姜黄素与顺铂在抑制人舌鳞癌Tca-8113细胞增殖时具有协同效应,其机制可能与上调Notch1信号通路、下调EGFR信号通路有关。
[Abstract]:Objective To observe the effect of curcumin or curcumin combined with cisplatin on human tongue squamous cell carcinoma (Tca-8113) cells in vitro, and to observe the effect of its effect on the proliferation of human tongue squamous cell carcinoma cells, and to explore the possible anti-tumor mechanism. Methods the cultured Tca-8113 cells were randomly divided into curcumin group, cisplatin group, curcumin combined with cisplatin group and control group.MTT method to detect each drug. The inhibitory effect on the proliferation of Tca-8113 cells in tongue squamous cell carcinoma; the effect of Hoechst 33258 staining on the morphology of nucleus of tongue squamous cell carcinoma; flow cytometry to detect the apoptosis rate; the effect of Western blotting on the expression of Notch1 and epidermal growth factor receptor (EGFR) protein in Tca-8113 cells of tongue squamous cell carcinoma. Results MTT method junction The effect of curcumin combined with cisplatin was significantly higher than the curcumin group and cisplatin group (P0.05). The results of flow cytometry showed that the apoptosis rate of curcumin combined with cisplatin group was significantly higher than that of curcumin group and cisplatin group (P0.05).Western blotting, and the ratio of curcumin combined with cisplatin group was compared with cisplatin group (P0.05). Compared with the curcumin group and cisplatin group, the expression of Notch1 was obviously up, and the expression of EGFR was obviously down. The effect of curcumin combined with cisplatin on the expression of Notch1 and EGFR was significantly different from that of the curcumin group and cisplatin group (P0.05). Conclusion curcumin and cisplatin have synergistic effects on the proliferation of Tca-8113 cells in human tongue squamous cell carcinoma. The mechanism may be related to up regulation of Notch1 signaling pathway and down regulation of EGFR signaling pathway.
