肺癌组织皮层肌动蛋白的表达及对肺癌细胞侵袭、迁移的影响

【摘要】：目的探讨皮层肌动蛋白(Cortactin)在肺癌组织的表达与患者生存预后的关系及对肺癌细胞侵袭、迁移的影响。方法收集贵州省人民医院50例行肺叶切除术的非小细胞肺癌(NSCLC)患者癌组织及癌旁组织,免疫组化染色检测肺癌组织Cortactin表达,分析Cortactin表达与肺癌患者临床资料的关系。对肺癌患者进行随访,记录患者5年生存率;以5年生存率作为评价指标,采用单变量和多变量Cox比例风险模型评价患者预后的影响因素。采用小干扰RNA(siRNA)技术抑制肺癌A549细胞Cortactin的表达,划痕实验检测细胞迁移能力,Transwell实验检测细胞侵袭能力。结果肺癌组织Cortactin高表达率为72.0%(36/50),癌旁组织高表达率为24.0%(12/50),肺癌组织Cortactin高表达率明显高于癌旁组织,两组间比较差异具有统计学意义(P0.01)。肿瘤最大径≥3cm组Cortactin高表达率明显高于肿瘤最大径3cm组,中高分化组Cortactin高表达率明显高于低分化组,有淋巴结转移组Cortactin高表达率明显高于无淋巴结转移组(均P0.05);Cortactin表达与性别、年龄、TNM分期、组织学类型无关(均P0.05)。Cortactin低表达组5年总生存率为42.1%,高表达组5年总生存率为13.7%,高表达组5年总生存率明显低于低表达组(P=0.018)。Cox多因素分析显示肿瘤直径大、淋巴结转移、高Cortactin表达是影响患者总生存期的独立危险因素(均P0.05)。划痕实验显示Cortactin-siRNA组细胞迁移率明显小于Cortactin-NC组、Cortactin-BC组;Transwell实验显示CortactinsiRNA组穿膜细胞数明显少于Cortactin-NC组、Cortactin-BC组,差异均具有统计学意义(均P0.05)。结论 Cortactin表达与肺癌患者生存预后有关,抑制Cortactin表达能够降低肺癌细胞的侵袭、迁移能力,Cortactin有望成为肺癌治疗的新靶点。
[Abstract]:Objective to investigate the relationship between the expression of cortical actin Cortacin and the survival and prognosis of patients with lung cancer and its effect on the invasion and migration of lung cancer cells. Methods the tissues and adjacent tissues of 50 patients with non-small cell lung cancer (NSCLC) undergoing lobectomy in Guizhou Provincial people's Hospital were collected. The expression of Cortactin in lung cancer tissues was detected by immunohistochemical staining. The relationship between the expression of Cortactin and the clinical data of patients with lung cancer was analyzed. The 5-year survival rate was recorded and univariate and multivariate Cox proportional risk models were used to evaluate the prognostic factors of lung cancer patients. Small interference RNA-siRNAs were used to inhibit the expression of Cortactin in lung cancer A549 cells. The migration ability of A549 cells was detected by scratch test and the invasion ability was detected by Transwell assay. Results the high expression rate of Cortactin in lung cancer tissues was 72.0 / 36 / 50 and that in adjacent tissues was 24.0 / 12 / 50. The high expression rate of Cortactin in lung cancer tissues was significantly higher than that in paracancerous tissues. The difference between the two groups was statistically significant (P 0.01).
