不饱和脂肪酸对人胃癌细胞蛋白激酶B信号通路的调控作用

【摘要】:目的研究不饱和脂肪酸对人胃癌细胞蛋白激酶B(PKB/Akt)信号通路的调控作用。方法取胃癌患者肿瘤组织,移植到重症联合免疫缺陷(SCID)小鼠上,将移植瘤组织酶解消化,分离出细胞进行传代培养。然后将传代细胞分为5组:低中高3个浓度实验组(不饱和脂肪酸0.3,0.6,1.2 mg·L~(-1)),对照组(5-氟尿嘧啶10 mg·L~(-1)),溶剂对照组(二甲基亚砜30μL·L~(-1)),给药后培养48 h。用噻唑蓝(MTT)法检测不饱和脂肪酸对人胃癌细胞的增殖抑制作用,用酶联免疫吸附法检测胃癌细胞中p21、双微体基因(MDM2)及PKB/Akt表达水平。结果高中低3个浓度实验组人胃癌细胞增殖抑制率比对照组分别升高了31.7%,55.51%,68.28%;这3组的p21和MDM2水平比对照组分别升高了4.39%,80.12%,161.36%和19.81%,97.76%,207.35%;这3组的PKB/Akt水平比对照组降低了13.31%,57.69%,78.71%。结论不饱和脂肪酸对人胃癌细胞增殖的抑制作用主要与升高肿瘤细胞内p21与MDM2水平和抑制PKB/Akt信号通路的活性有关。
[Abstract]:Objective to investigate the regulatory effect of unsaturated fatty acids on protein kinase BPKB / Akt signaling pathway in human gastric cancer cells. Methods the tumor tissues of gastric cancer patients were transplanted to mice with severe combined immunodeficiency syndrome (SCID). The transplanted tumor tissues were digested by enzyme and the cells were isolated and cultured. Then the passage cells were divided into five groups: low, medium and high concentration experimental group (unsaturated fatty acid 0.3U 0.61.2mg / L), control group (10 mg / L 5-fluorouracil) and solvent control group (30 渭 L / L DMSO) cultured for 48 h. The inhibitory effect of unsaturated fatty acids on the proliferation of human gastric cancer cells was detected by thiazolyl methacrylate (MTT) assay. The expression levels of p21, MDM2 and PKB / Akt in gastric cancer cells were detected by enzyme-linked immunosorbent assay (Elisa). Results compared with the control group, the inhibition rate of proliferation of gastric cancer cells in the three experimental groups increased 31.75.51 and 68.28.The levels of p21 and MDM2 in the three groups were 4.3980.120.12and 97.767.36% higher than those in the control group, respectively, and the PKB / Akt level in these three groups was 13.31% lower than that in the control group (78.71%). Conclusion the inhibitory effect of unsaturated fatty acids on the proliferation of human gastric cancer cells is related to the increase of p21 and MDM2 levels and the inhibition of PKB / Akt signaling pathway activity.
