miR-125b-1-3p在胃癌中的表达及意义

【摘要】:目的:本研究通过探索miR-125b-1-3p在胃癌及对应癌旁组织中的表达情况,并分析其与胃癌临床病理特征之间的关系,从而探讨miR-125b-1-3p作为胃癌诊断标志物和治疗靶点的可能性。方法:实验前期利用基因芯片技术检测14例早癌患者的胃癌组织及对应癌旁组织中miRNAs的表达谱分析情况。收集42例手术患者的胃癌组织及对应的癌旁组织。首先在胃癌样本及癌旁样本中提取总的RNA,运用实时荧光定量PCR检测出miR-125b-1-3p在胃癌组织中表达水平,并分析miR-125b-1-3p的表达水平与胃癌临床病理特征之间的关系。结果:实时荧光定量PCR结果提示胃癌组织miR-125b-1-3p的表达相较于对应癌旁组织存在显著下调,有统计学差异。miR-125b-1-3p在胃癌的低表达仅与分化程度存在相关性,而与性别、TNM分期、浸润深度、淋巴结转移、年龄、肿瘤大小等胃癌临床病理特征均无明显相关性。在miR-125b-1-3p低表达的胃癌组织中,根据ROC曲线确定胃癌组织高/中分化程度的曲线下面积是0.714(P0.05),得出高/中分化程度的最佳界值:相对表达量为0.18,其在诊断高/中分化程度的敏感度70%、特异性72.2%、阳性预测值70%、阴性预测值30%。当相对表达量≥0.18时发生高/中分化程度的风险是相对表达量0.18时患者的2.33倍(P0.05),提示相对表达量≥0.18是胃癌组织高/中分化程度的独立预测因素。结论:miR-125b-1-3p在胃癌组织中显著下调,当miR-125b-1-3p的相对表达量≥0.18在预测胃癌高/中分化程度上有较高的诊断价值。
[Abstract]:Objective: to explore the expression of miR-125b-1-3p in gastric cancer and its relationship with the clinicopathological features of gastric cancer, and to explore the possibility of miR-125b-1-3p as a diagnostic marker and a therapeutic target for gastric cancer. Methods: the expression profiles of miRNAs in gastric cancer tissues and adjacent tissues of 14 patients with early cancer were detected by microarray technique. Gastric cancer tissues and corresponding paracancerous tissues were collected from 42 patients. First, the total RNAs were extracted from gastric cancer samples and adjacent samples. The expression of miR-125b-1-3p in gastric cancer tissues was detected by real-time fluorescent quantitative PCR, and the relationship between the expression level of miR-125b-1-3p and clinicopathological features of gastric cancer was analyzed. Results: the results of real-time fluorescence quantitative PCR showed that the expression of miR-125b-1-3p in gastric cancer was significantly down-regulated compared with the corresponding adjacent tissues, and the low expression of miR-125b-1-3p in gastric cancer was only correlated with the degree of differentiation, but with the TNM stage and depth of invasion. The clinicopathological features of gastric cancer, such as lymph node metastasis, age and tumor size, were not significantly correlated. In gastric cancer tissues with low expression of miR-125b-1-3p, According to the ROC curve, the area under the curve of high / medium differentiation degree of gastric cancer is 0.714 (P0.05). The best boundary value of high / medium differentiation degree is obtained: the relative expression is 0.18, its sensitivity in diagnosing high / medium differentiation degree is 70, specificity 72.2, positive.
