Hedgehog信号通路阻断剂GANT61对胃癌细胞株MC的抑制作用

【摘要】:目的:探讨Hedgehog信号通路阻断剂GANT61对胃癌细胞株MC的细胞毒性及其对MC凋亡和迁移能力的影响。方法:采用不同浓度的GANT61处理胃癌细胞MC后,MTT法检测GANT61对胃癌细胞株MC的细胞毒性;流式细胞术检测GANT61对MC细胞周期以及凋亡的影响;Western blot验证GANT61对胃癌细胞株MC中Gli1蛋白表达的影响;细胞划痕实验检测GANT61对胃癌细胞株MC迁移能力的影响。结果:采用GANT61浓度20umol/L处理细胞时,细胞活性下降50%,MC细胞凋亡率增加(实验组6.60±1.04%vs对照组2.47±0.47%);GANT61能够下调胃癌细胞株MC中Gli1蛋白的表达,另外GANT61能有效抑制胃癌细胞株MC的迁移能力。结论:GANT61通过阻断Hedgehog信号通路,表现出显著的细胞毒性,有效抑制胃癌细胞株MC的迁移能力并诱导胃癌细胞株MC的细胞周期阻滞及细胞凋亡,为胃癌的靶向治疗提供一种新的治疗选择与思路。
[Abstract]:Aim: to investigate the cytotoxicity of Hedgehog signaling blocker GANT61 on gastric cancer cell line MC and its effect on apoptosis and migration. Methods: the cytotoxicity of GANT61 to gastric cancer cell line MC was detected by MTT assay after treated with different concentrations of GANT61. The effect of GANT61 on cell cycle and apoptosis was detected by flow cytometry. Western blot was used to test the effect of GANT61 on the expression of Gli1 protein in gastric cancer cell line MC, and the effect of GANT61 on the migration ability of gastric cancer cell line was detected by cell scratch assay. Results: when treated with 20 umol / L GANT61, the cell activity decreased 50% and the apoptosis rate of MC increased (experimental group 6.60 卤1.04%vs 2.47 卤0.47%). GANT61 could down-regulate the expression of Gli1 protein in gastric cancer cell line MC, and GANT61 could effectively inhibit the migration ability of gastric cancer cell line MC. Conclusion by blocking the Hedgehog signaling pathway, the cell cycle arrest and apoptosis of gastric cancer cell line MC can be effectively inhibited. To provide a new therapeutic choice and thinking for the targeted treatment of gastric cancer.
