乳腺癌新辅助化疗前后p62和XIAP表达的变化及其意义

【摘要】:目的探讨乳腺癌新辅助化疗(neoadjuvant chemotherapy,NCT)前后自噬的变化及其与XIAP变化的关系。方法应用免疫组化En Vision两步法检测NCT前后94对乳腺癌组织中p62和XIAP的表达,观察NCT后p62和XIAP表达的变化情况,分析其相关性。结果 NCT前乳腺癌标本中,p62高表达率为51.1%(48/94),XIAP高表达率为84.0%(79/94);NCT后乳腺癌标本中,p62高表达率升高为81.9%(77/94),XIAP高表达率降低为35.1%(33/94)。即NCT后p62高表达率明显升高,XIAP高表达率明显降低(P均0.05)。NCT前后相比,p62表达升高的样本率为73.4%,XIAP表达下降的样本率为75.5%,且p62表达变化与XIAP表达变化呈显著负相关(P0.01)。结论乳腺癌NCT后自噬增强与XIAP下调共存,且XIAP可能参与NCT后自噬增强的负性调节。
[Abstract]:Objective to investigate the changes of autophagy in breast cancer before and after neoadjuvant chemotherapy (neoadjuvant) and the relationship between autophagy and XIAP. Methods the expression of p62 and XIAP in 94 cases of breast cancer before and after NCT was detected by immunohistochemical en Vision two-step method. The expression of p62 and XIAP was observed after NCT, and the correlation between p62 and XIAP was analyzed. Results the high expression rate of p62 in breast cancer specimens before NCT was 51.1% (48 / 94). The high expression rate of XIAP was 84.0% (79 / 94). The high expression rate of p62 was increased to 81.9% (77 / 94) and decreased to 35.1% (33 / 94). That is to say, the high expression rate of p62 was significantly increased after NCT. The high expression rate of XIAP decreased significantly (P0. 05). The sample rate of increased p62 expression before and after NCT was 73.5%, and the change of p62 expression was negatively correlated with the change of XIAP expression (P0.01). Conclusion autophagy after NCT in breast cancer coexists with down-regulation of XIAP, and XIAP may participate in the negative regulation of autophagy after NCT.
